ObjectiveTo investigate the protection of jiangyabaolan tablet on traditional Chinese medicine against target organ damage in spontaneously hypertensive rats(SHR), primarily explore the mechanism of left ventricular hypertrophy,renal damage from ACE-AngⅡ-AT1 R for clinical application.MethodsAccording to the weight and blood pressure,SHR were assigned randomly into 3 groups, the SHR control group and Enalapril group and jiangyabaolan tablet group, Wistar-kyoto(WKY)-rats were used as negative control group; 10 per set.After gavage for 8 weeks, by using noninvasive sphygmomanometer to measure last arterial blood pressure,such as SBP,DBP,MBP, before the medication,medication after 3d and ervery other week when rats are awake; weighed heart and left ventricular from rats to calculate cardiac index(CI), left ventricular index(LVMI) and renal index(RI), observed pathological changes in left ventricular under light microscope stained with HE, using pathological image analysis system to determine cardiomyocytes transection diameter(CTD); Enzyme-linked immunosorbent assay(ELISA) to detect myocardial angiotensin Ⅱ(Ang Ⅱ) and angiotensin-converting enzyme(ACE),Western blot detects the expression of myocardial protein angiotensin Ⅱ type 1 receptor(AT1R).Result1. Effect of jiangyabaolan tablet on SHR blood pressure, heart rate.Compared with WKY group,SBP,DBP,and MBP on SHR in SHR group weresignificantly increased(P<0.01),HR has a tendency to increase without statistical significance(P>0.05);jiangyabaolan tablet can significantly reduce the SBP,DBP,and MBP of SHR,but significantly lower than postitive control Enalapril;had no obvious effect on HR(P>0.05).2.Jiangyabaolan tablet influence on SHR cardiac index,left ventricular indices,renal indexCompared with WKY group, the CI, LVMI of SHR rats increased significantly(P<0.01). Jiangyabaolan tablet can significantly reduce CI, LVMI,had significant difference with SHR control group(P<0.05).3. Pathological effects on SHR left ventricular myocardial cell diameter and the influence of the pathogogic morphologyCompared with WKY group, left ventricular myocardial cell diameter was significantly increase(P<0.01), part of the cardiomyocytes hypertrophy under the microscope, myocardial fiber swelling degeneration, disorganized, broken and could see the infiltration of inflammatory cells. Jiangyabaolan tablets could significantly improve myocardial pathological degrees(P<0.05).4. Effects of jiangyabaolan tablet treasure ACE-AngⅡ-AT1 R in SHR left ventricularCompared with WKY group, AT1 R protein expression of SHR group was significantly enhanced(P<0.01);left ventricular myocardial tissue ACE activity and localized content AngⅡshowed a downward trend without statistical significance(P>0.05).AT1 R protein expression was significantly down-regulated by Jiangyabaolan tablet on SHR(P<0.05),but had no significant effect on ACE and AngⅡ(P>0.05).5. Effects of jiangyabaolan tablet treasure ACE-AngⅡ-AT1 R in SHR renal tissueCompared with WKY group, AT1 R protein expression of SHR renal tissue showed a upward trend(P<0.05), ACE activity and localized content AngⅡshowed a upward trend without statistical significance(P>0.05).Jiangyabaolan tablet can significantly reduce the activity of ACE,AngⅡ content(P<0.01), and lower AT1 R protein expression(P<0.05).Conclusion1. jiangyabaolan tablet can lower SHR blood pressure.2. The 20-week-old SHRs have obvious pathological changes, such as left ventricular hypertrophy,renal damage.3. Jiangyabaolan tablet has varying degrees prevention and reversal effect left ventricularhypertrophy,renal damage in rats with hypertension.4. jiangyabaolan tablet piece mechanism of anti hypertensive left ventricular hypertrophy and renal damage are related to the inhibition of target organs’ ACE-AngⅡ-AT1 R.
