Objective To analyze the biochemical index, variation of coaulation and cytokines ofrats with exertional heat stroke(EHS),observe the effect of heparin on the earlyintervention of EHS rats and to discuss the role. Methods20male Sprague-Dawleyrats were divided into two groups to establish EHS models.We used heat stresscomplicated with high intensity exercise to make EHS happen and examined theirbiochemical index (CK, Cr, AST, ALT) to prove the scientificity. Then other60maleSprague-Dawley rats were randomly assigned to the sham group (Group A,n=8),exertional heat stroke with saline group (Group B,n=26), or heparin-treatedexertional heat stroke group (Group C,n=26). Establish the rat models of EHS by theway of heat stress complicated with exercise from Group B and Group C. Afterestablishing the EHS rat models firstly we made survival analysis with10ratsrandomly from Group B and Group C (Group B1and Group C1). The remainingGroup B and Group C rats were randomly divided into two subgroups (Group B2,B3,Group C2,C3, n=8) and were examined the levels of blood platelet(PLT), activatedpartial thromboplastin time(APTT), prothrombin time(PT), D-Dimer, and fibrinogen(Fib) according to the following time points:0h (just prior to heat stress) and2hoursafter the onset of EHS. Meanwhile serumInterleukin-1β (IL-1β), Interleukin-6(IL-6),tumor necrosis factor-α (TNF-α), and high mobility group protein (HMGB1) contentwere measured by enzyme-linked immunosorbent assay (ELISA). The rats werekilled at the time and we took out of their intestinals which was then fixed by4%formalin. The Paraffin seetion of intestinal was dyed by HE. The pathologic changesof intestinal were abserved by light microseope. Results Eight-hour mortalityimproved in Group C1versus Group B1(P <0.05). Compared with Group A, Group B2,B3,C2and C3rats showed significant systemic infection reaction and activitedcoagulation. Compared with Group A, Group B2, B3and C3rats also showedincrease in serum TNF-α, IL-1β, IL-6, HMGB1(P <0.05). But their content ofGroup C2and Group C3were lower than that of Group B at the same time (P <0.05).Meanwhile, multiple tissue ischemia and injury happened. Conclusion (1)We canestablish EHS rat models with treadmill running in the hot laboratory.(2)Heparinmight improve the motality of EHS rats.(3) Heparin using before heat stress maylessen the intestinal injury and protect related organs by ameliorating excessivesystemic inflammation and hypercoagulable state.
