Objective:To observe the changes of mRNA and proteinexpression of cell matrix metalloproteinase2、matrix metalloproteinase9、Tissue Inhibitor of Metalloproteinase2(TIMP2)、CyclinD1in cervicalcancer cells stimulated by baicalein.Methods: Hela cells were cultured invitro and were exposed to baicalein(0,50,100,200and300)μM for(12,24,48)h.The periodic changes of each group cell were detected byflow cytometry,then the activity changes of each group cell weredetected by Gelatin Zymography,RT-PCR was used to measure themRNA changes of MMP2/9、TIMP2、CyclinD1and Western Blot wasused to measure the protein expression of MMP2/9、 TIMP2、CyclinD1.Results: Compared to control group, baicalein significantlyblocked cell cycle G1and decreased the activity of MMP2/9,decreasedthe mRNA and protein expression of MMP2/9、 CyclinD1in aconcentration-dependent manner (P<0.05,P<0.01). The mRNA andprotein expression of TIMP2significantly increased (P<0.05,P<0.01).Conclusion:Baicalein induced HeLa cell growth inhibition byblocked cell cycle G1and decreased the activity of MMP2/9,down-regulated the mRNA and protein expression of MMP2/9、CyclinD1,by up-regulated the mRNA and protein expression of TIMP2.
