OBJECTIVES:To investigate the effects of azithromycin on gastric emptying and plasma motilin in mouse, thus exploring mechanism of azithromycin to Gastrointestinal Symptom.METHODS:Eighty wistar rats were randomly dividing in10groups, i.e., the normal control group, small doses1-day group, small doses3-day group, small doses6-day group, normal doses-day group, normal doses3-day group, normal doses6-day group, large doses1-day group, large doses3-day group, large doses6-day group, and8rats in each group. The rats of observer groups were gastrogavaged by azithromycin. The dosages of small, normal and large groups were3mg/100g,6mg/100g and12mg/100g, respectively. First, gastric emptying was measured in unanesthetized rats by single photon emission computed tomography (SPECT) after gavaged milk loaded with technetium-99m diethylenetriamine pentaacetic acid(99mTc-DTPA), the gastric half-emptying time were calculated. Subsequently, blood was collected from the abdominal aorta to measure plasma motilin.RESULTS:In comparison to rats in control group,GET1/2decreased significantly in observer groups. The concentrations of plasma motilin in normal and large groups increased significantly, comparing with control group. However, there was no significant difference in small dose group.CONCLUTION:Azithromycin could promote gastric emptying. The mechanism of it not only are motilin-like effection, but also the participation of endogenous motilin.
