Objective: Evaluating Cnidii Fructus hypnotic active constituent (CHC) onsleep-deprived rats hypnosis and promote the learning and memory effects. Investigatingthe mechanism of CHC hypnosis and memory facilitationMethod: By intraperitoneal injection of PCPA (para-chlorophenylalanine)combined with small platform water environment to establish the rat sleep deprivationmodel. At the same time, gastric perfusion of high dose of CHC (160mg/kg), mediumdose of CHC (80mg/kg), low dose of CHC (40mg/kg),6consecutive days, and set aDiazepam positive control group (3mg/kg). Observing CHC on the synergistic hypnoticdose of pentobarbital sodium of sleep latency, sleep lasting time. Evaluating thehypnogenesis of CHC. Morris water maze test evaluation of spatial cognition andmemory ability in rats. The contents of5-HT, NA, GABA and5-HT, GABA brainstemin plasma was tested by ELISA, NA content in brain tissues by high performance liquidchromatography detection, analysis the effect of CHC on sleep deprivation studyneurotransmitter content related to memory and sleep in blood and brain tissue in rats.Results:1. Effect of CHC sodium pentobarbital on sleep deprivation rat synergistichypnotic: The sleep latency of CHC high, medium dose group was significantly lowerthan the model group (P<0.001); sleep lasting time CHC high, medium dose group weresignificantly higher than the model group (P<0.001, P<0.01).2. Effect of CHC on sleep deprivation rat MWM spatial cognition and learningand memory: In place navigation test, the escape latency of CHC high dose group wassignificantly lower than the model group (P<0.05); search strategy model group, mostlyedge type, CHC medium dose group and blank control group, mostly straight or trendtype, and model group were significantly different (P<0.05). In spatial probe test, targettime CHC medium dose group was significantly higher than the model group (P<0.01), through time CHC high, medium dose group were significantly lower than the modelgroup (P<0.05), times across the high dose group was significantly higher than themodel group (P<0.05); exploration strategy model group, mostly edge type, CHC high,medium dose group and blank control group, mostly trend type, and model group weresignificantly different (P<0.01, P<0.05).3. Effect of CHC on sleep deprivation rats’ neurotransmitter in the plasma andbrain stem: There is no significant difference in plasma5-HT, NA, GABA contentamong groups. No significant differences in brain stem5-HT, GABA content amonggroups; The content of NA, the model control group was significantly higher than that inthe control group (P<0.01), CHC of high dose and middle dose group was significantlylower than the model control group(P<0.05)。Conclusion: CHC has significant hypnotic effect on the sleep-deprived rats ofPCPA small platform water environment, and significantly promote their spatialcognition and spatial reference memory. The mechanism of action may changethe correlation with the content of neurotransmitter in brain stem.
