Supercritical liquid chromatography(SFC) is a kind of chromatography technology developed in 1980 s, with supercritical fluid as mobile phase. The density of supercritical fluid is similar to fluid. The viscosity of supercritical fluid is similar to gas. The diffusion coefficient is much higher than other liquid. So, it is quickly in mass transfer, it can reach equilibrium with short time and higher efficient separation. As the temperature of CO2 close to room temperature, green non-toxic non flammable, no chemical corrosion, thus it become the first choice. So, the SFC has some advances, for example, high separation efficiency, short separation time and so on. With the continuous improvement of the technology and theory, the SFC has caused the attention of the separation researchers.The traditional Chinese medicine with a long history of five thousand years, is our country's national treasures. Our country also is regarded as "kingdom of traditional Chinese medicine", and has realized the modernization of the separation and purification of Chinese medicine effective component technology, is the path to the development of the traditional Chinese medicine enterprise. Using modern advanced science and technology to extracte and purify the active ingredients, clarify its base of curative effect, improved traditional Chinese medicinal materials and the preparation of standard system for the development of traditional Chinese medicine industry has profound significance. At present, the application of SFC with the separation and purification of effective components in Chinese medicine has achieved satisfactory results.In this paper, the SFC conditions were established ahout the effective components of traditional Chinese medicine Fructus Cnidii, Fructus Psoraleae, Magnolia officinalis, and Polygonum cuspidatum selection and optimization. Using HPLC to detect the purity, the chemical structure of target compound were identified by ultraviolet spectrum(UV),nuclear magnetic resonance(NMR) and mass spectrometry(MS).1. Using SFC to isolate and purify two coumarins in Fructus Cnidii.This paper has established the SFC method of purification of effective components infructus cnidii. Using ethanol and ether to extract the effective components of Fructus Cnidii, through SFC to separation and purificationin, in this process, studying the effection of chromatographic column, modifier types and proportion, mobile phase flow rate,pressure and temperature to the capacity factor and the degree of separation. Determine the optimal conditions for SFC separation of osthole and imperatorin, the structure of osthole and imperatorin were authenticated. According to the components' chemical structure and the order in separation process, ensure, under the above chromatographic conditions SFC separation process for normal phase chromatography process. The thermodynamics laws of SFC separation process were studied, through comparing the enthalpy and entropy variations in the phase transfer process of osthole and imperatorin, the SFC separation process is controlled by enthalpy.2. Using SFC to isolate and purify two coumarins in Fructus Psoraleae.This paper has established the SFC method of purify effective components from the Fructus Psoraleae. Using SFC to isolate and purify two Magnolia lignin composition.This paper established the SFC method of purification active ingredients in magnolia bark. Using methanol and ether to extract the effective components of Fructus Psoraleae.through SFC to separation and purificationin, in this process, studying the effection of chromatographic column, modifier types and proportion, mobile phase flow rate, pressureand temperature to the capacity factor and the degree of separation. Determine the optimal conditions for SFC separation of magnolol and honokiol. The structure of magnolol and honokiol were authenticated. According to the components' chemical structure and the order in separation process, ensure, under the above chromatographic conditions SFC separation process for normal phase chromatography process.4. Using SFC to isolate and purify two anthraquinones in Polygonum cuspidatum.This paper established the SFC method of purification effective components in the Polygonum cuspidatum.Using methanol and ether to extract the effective components.Then through SFC to separation and purificationin, in this process, studying the effection of chromatographic column, modifier types and proportion, mobile phase flow rate,pressure and temperature to the capacity factor and the degree of separation. Determine the SFC optimal conditions of separation physcion and emodin. According to the components' chemical structure and the order in separation process, ensure, under the above chromatographic conditions SFC separation process for normal phase chromatography process. The thermodynamics laws of SFC separation process were studied, through comparing the enthalpy and entropy variations in the phase transfer process of physcion and emodin, the SFC separation process is controlled by enthalpy.
