Objective:This research in view of "salty into kidney" from neijing, combined with modern scientific research, Proposed salt-sensitive hypertension and kidney disease located in the hypothesis. Enriched the pathogenesis theory of salt-sensitive hypertension in traditional Chinese medicine; Combined with the current salt-sensitive hypertension latest genomics research, Proposed to explore the molecular mechanisms of salt-sensitive hypertension from kidney-specific expression of the molecule scientific hypothesis; In order to verify these hypothesis,we study the expression of kidney-specific cadherin, And the depth study the mechanisms of KSP abnormal expression leads to salt-sensitive hypertension. Thus proposed a new molecular mechanism of salt-sensitive hypertension,and also expanded the theory of "salt into kidney"in traditional Chinese medicine.Method:Experimental studies on kidney-specific cadherin expression of the salt-sensive rat.Experiments adopt DS rats created by Dahl, Randomly divided into three groups, Model+drug group,Model group,Blank group. In five weeks,Cast model group to8%salt diet, Cast model+drug group to8%salt diet and traditional Chinese medicine kidney Recipe(Include Eucommia, mistletoe, Achyranthes, wolfberry).Give the blank group normal diet.Observed the changes in blood pressure. Experimental fifth week(Began to cast high-salt diet for the first day of the experiment),kill the rats, Take Urine, Each group of rats was measured in NAG. Take blood, Measured SCR, BUN, TP, ALB and other biochemical markers; Take kidneys.calculation the organ index, HE staining in histological changes. Mouse anti-Ksp-Cadherin kit for measuring kidney-specific cadherin expression in kidney.Results:After the modeling, Blood pressure of the model group higher than the model+drug group and blank group (P<0.05), model+drug group and blank group, no significant difference in blood pressure (P>0.05), Blood urea nitrogen (BUN) model group was significantly different from the model+drug group and blank group (P<0.05),No significant differences in the model+drug group and blank group (P>0.05); No significant difference between the three groups in serum creatinine; Tp numerical blank group compare with the model group and the model+drug group have Significant differences (P<0.05). Alb numerical blank group and model+drug group was statistically significant (P<0.05); Urinary protein and urinary NAG numerical model group was significantly different from the model+drug group and the blank group (P <0.05), No significant differences in the model+drug group and blank group (P>0.05); Organ index of rats in each group had no significant difference (P>0.05);The ksp model group was significantly different from the model+drug group and the blank group (P <0.05), No significant differences in the model+drug group and blank group (P>0.05)Conclusion:1. Traditional Chinese medicine herbs on early intervention have a role in salt-sensitive hypertension2.The model group was significantly worse than the model+drug group and blank group in kidney damage.This damage from the renal tubules.3. High-salt diet caused the abnormal expression of ksp, we can verify the proposed assumptions:High-salt diet caused the abnormal expression of ksp, Thus forming tubulointerstitial injury Resulting in salt-sensitive hypertension.
