Expression And Significance Of SOX2、OCT4、TWIST And SNAIL Protein In Colorectal Carcinoma

Objective: To study the expression and significance of SOX2, OCT4, TWIST andSNAIL protein in colorectal carcinoma and its correlation with clinicopathologic featuresin colorectal carcinoma.Methods: In115cases of invasive colorectal carcinoma，75cases of colorectaladenoma and35cases of colorectal musocal tissue, the expression of SOX2, OCT4,TWIST and SNAIL proteins were determined by the immunohistochemical staining,respectively. The relationship between these three proteins and clinicopathologic featuresin colorectal carcinoma was analyzed by χ2test and Spearman’s rank correlation analysis.The survival analysis was analyzed by Kaplan-Meier’s method and Cox regressionmethod.Results:(1) In115cases of invasive colorectal carcinoma, the positive rates of SOX2,OCT4, TWIST and SNAIL protein were53.9%,58.3%,67.0%,71.3%, respectively(p<0.05).(2) Immunostaining showed more SOX2positive cells in patients with advancedN/TNM stages, or patients with distant metastasis, or advanced TNM stages than that inlower N/TNM stages, or patients without distant metastasis, or lower TNM stage(p<0.05).And also immunostaining showed more OCT4positive cells in patients with distantmetastasis, or advanced TNM stage than that in patients without distant metastasis, orlower TNM stage (p<0.05).(3) Spearman’s rank correlation analysis shown SOX2correlated with TWIST and SNAIL positively，and OCT4correlated with SNAIL positively,and TWIST correlated with SNAIL positively. And OCT4did not correlate with SOX2andTWIST.(4) Kaplan-Meier’s methods analysis shown median survival time of SOX2,OCT4, and TWIST positive expression was shorter than that of negative expression (p＜0.05). The5-year survival rate of SOX2, OCT4, and TWIST positive expression was lower than that of negative expression (p＜0.05). Cox regression analysis shown TWIST positiveexpression and LN metastasis were independent prognostic index.Conclusion:(1)SOX2and OCT4testing could predict the invasive behavior andhigher clinical TNM stages of colorectal carcinoma;(2) The over-expression of SOX2,OCT4and TWIST correlates with the metastasis of colorectal carcinoma and unfavorableprognosis.
