Study On Application And Evaluation Of Taste-masking Technology Taking Andrographis As A Model

The drug taste effects compliance of patients. Many drugs are too bitter to swallow, restricted in formulation development and clinical use. Meanwhile bitter drugs can also cause great psychological and physical burden to patients, especially children, the elderly and patients with poor appetite, and could easily lead to refuse service and vomiting. The ill taste problem of traditional Chinese medicine is especially serious, most traditional Chinese medicine have strong bad taste. Therefore, this thesis taked application of taste-masking technology in traditional Chinese medicine as the object.The bitter medicine Andrographis was selected as the model drug. By the molding process selection and optimization, which was made into granules. Then add flavoring agents, using volunteers tasting drugs and the electronic tongue analysing drugs to evaluate taste-masking effects. Screen out the better taste-masking prescriptions to improvement or hide Andrographis bitter. The main contents are as follows:With the index of granules formability, moisture absorption, particle state, with or without caking, bulk density, angle of repose, melting resistance, hardness, size distribution and so on, the formation technique involving the sorts and the proportion of the additives, wetting agent concentration was determined by single factor test. The results determine the selection of the molding process as Andrographis particulate materials was dextrin, and the ratio of Andrographis dry powder and dextrin was1:2, wetting agent was75%ethanol.On this basis, add flavoring agents, study on the taste-masking of Andrographis granules. Volunteers tasted drugs and evaluated taste-masking effects, using comprehensive score method to screen out three better taste-masking prescriptions. The best prescription was the ratio of Andrographis dry powder and dextrin1:2, APM0.3%, AK0.2%, HPMC k4M1.0%, citric acid0.8%, NaCl1.0%. Using the electronic tongue to analyzed the three better taste-masking prescriptions, the results were consistent with the results of Volunteers tasted drugs.Define the quality standards of Andrographis extract and granules, including character, TLC, HPLC content of andrographolide and dehydroandrographolide and other projects.
