Background and ObjectiveEsophageal cancer ranks the eighth among worldwide malignancies with the fatality rate ranking the sixth. Pathological types of esophageal cancer contain adenocarcinoma and squamous cell carcinoma. The main pathological type in our country is esophageal squamous cell carcinoma (ESCC) which has un-conspicuous early clinical features. Surgical treatment on this kind of caner will lead to great bodily injury and very low survival rate for the patients. Therefore, esophageal carcinoma is a serious threat to human health.Pituitary tumor-transforming gene1(PTTG1) is isolated from rat pituitary tumor and it has been considered as the key pituitary tumor-transforming gene. PTTG1participates in the formation process of tumors by inducing cell transformation and promoting the expression of other proto-oncogene and promoting tumor factors. Studies have shown that PTTG1has been expressed in many malignancies including esophageal cancer,participated in the differentiation and metastasis of tumor cells and played an important role in tumor progress.MicroRNA (miRNA) is the precursor of miRNA that contains stem-loop structure, and after Dicer processing, the,total length of a class of non-coding small RNA molecules consists of20-22nucleotides. miRNA has the functions of restraining the transcription, translation and shearing of targeted mRNA. In addition, it can regulate cell growth, histological differentiation and participate in the development and disease processes in life. Studies have shown that there is a phenomenon of abnormal expression of miRNA in malignant tumor, indicating that miRNA is closely related to the development of esophageal carcinoma.For this purpose, we selected61ESCC tissues and their normal esophageal squamous epithelial tissues as experiment specimens, detected the expression of PTTG1and miR-655in specimens and analyzed the relationship between the expression levels of PTTG1and miR-655and patients’ clinicopathologic features and survival time.Methods61cases of ESCC tissues and their normal esophageal squamous epithelial tissues were used as experiment specimens and the immunohistochemistry technique, RT-PCR technology and qRT-PCR technology were used to examine the expression of PTTG1protein, the relative expression of PTTG1mRNA and the relative expression of miR-655respectively. SPSS17.0statistical software was used to analyze the expression levels of PTTG1and miR-655as well as their correlation with clinicopathologic features and survival time.ResultsThe expression of PTTG1protein and PTTG1mRNA were significantly higer in ESCC than in corresponding normal tissues (P<0.01). The expression of miR-655was significantly lower in ESCC than in corresponding normal tissues (P<0.01). PTTG1and miR-655have significant negative correlation (r=-0.52,P=0.00). The expression of PTTG1protein、PTTG1; mRNA and miR-655were significant associated with lymph node metastssis、grade of histological differentiation and TNM stage,miR-655was also significant associated with tumor location(P<0.05). PTTG1protein and PTTG1mRNA were have none associated with tumor location(P>0.05).ConclusionsThe expression of PTTG1was rise abnormally and the expression of miR-655was lower abnormally in ESCC. miR-655may be involved the gene expression of PTTG1.
