Study Of Four Kinds Of Traditional Chinese Medicine Preparation In Rats Vivo Metabolism

Traditional Chinese medicine (TCM) clinical applications have several thousand years of history, but the use of modern medicine method of TCM research mechanism began in the1920s.Through the study of Chinese medicine pharmacology, not only can very good validation of TCM theory, but also to deepen and developing traditional Chinese medicine efficacy, make it more reasonable safe clinical applications have very important significance. Study on drug metabolism plays an important role in drug research and development.Recent years, metabolite analysis for chemical constituents of traditional Chinese medicine also atracts a great interest.Metabolite analysis of traditional Chinese medicine is a guid line to study the drug mechanism, develop new drugs, control medicine quanlity and help the use of clinic drugs. It has become an important cut-over point in the modernization process of traditional Chinese medicine.Modem TCM preparations are embodiments of the modernization of TCM. However, in order to modernize TCM preparations, it is more important to modernize the theoretical basis of dosage forms selected and the method for screening prescriptions, optimizing technics and evaluating quality than only to modernize pharmaceuical technique. Pharmacokinetics and pharmacology are subjects focusing on studying the regularities of drug’s absorption, distribution, metabolism and elimination in vivo, and further elucidating the relationships between preparational factors, biological factors and drug effects.HPLC is a powerful technology for drug research and develops rapidly recent several years.This method is rapid, high sensitive, accurate and specific, especially suitble to analyze strong polarity, non-volatile, high molecular weight, heat-instability samples.In the past years, it has greatly promoted the development of chug research.The application of HPLC to metabolite analysis of traditional Chinese medicine has significence in both theroy and practice.In this paper, We have carried out some research on three kinds of traditional Chinese Medicine:(Xuesaitong preparations, Dragon’s blood capsules, Berberine Hydrochloride Tablets and Fibrauretine Tablets).The main research components were panax notoginseng saponins、loureirin A and loureirin B、palmatine hydrochloride and berberine hydrochloride in three kinds of TCM, respectively. We used high performance liquid chromatography (HPLC) to three kinds of TCM after rats oral administration, and the main metabolites, in vivo pharmacology, sample pretreatment methods etc.. detailed work as follows:1. Quantitative determination of Xuesaitong preparations major panax notoginseng saponinsin rat plasma and brain after oral administration by HPLC/DAD (High Performance Liquid Chromatography) method;2. A rapid centrifugation assisted CPC-HPLC-DAD (Column Partition Chromatography-High Performance Liquid Chromatography) method for determination of loureirin A and loureirin B of Dragon’s blood capsules in rat plasma and urine after oral administration;3. Simultaneous quantification of berberine hydrochloride tablets and of fibrauretine tablets in rat urine、feces、bile and plasma by HPLC/DAD method and their pharmacological differences study。...
