| Background:Myocardial infarction is one of the serious fatal disease in critical condition. Though modern science provide a new idea of interventional therapy, which make survival rised,but the radical solution should be focus on preventive treatment.This research is based on the cardiac autonomic neural remodeling theory, definite quantitative determination of THã€AchEã€NGFã€CA,test homodynamic in molecular biology level. As for Investigate TRADITIONAL MEDICAL Formulae’s effect on cardiac autonomic neural remodeling, look forward to regulate autonomic nervous system activity, inhibition of malignant arrhythmia, improve the prognosis of patients with myocardial infarction. It’s a innovative path to focus on cardiac autonomic neural remodeling, play the advantages of TCM’s treating principles to prevent disease theories.Objective:To observe the acetyl cholinesterase (AchE) activity changes in myocardium after using Ningxia formula on the acute phase of myocardial infarction.Method:A total of86male SD rats were used for the experiments. Myocardial infarction model was prepared by ligation of the left anterior descending coronary artery and the sham operation rat was only braided without ligation. According to the method of random digits table, survival rats were divided into sham group and30-day groupã€Metoprolol groupã€Chinese high-dose group and traditional dose group.Sham group and30d group were given saline by gavage for30days, other groups were fed corresponding drugs by gavage for30days. The activities of TH and Ache were examined in myocardium at peri-infarcted area by the method of RT-PCR, and a quality identification of CA from plasma in order to investigate the role of Berberine-pinellia formula on the acute phase of myocardial infarction.Results:â‘ in myocardial infarction, the AchE mRNA levels in model group compared to the sham group was significantly increased, there was a significant difference (P<0.01)[MI (4.58±1.33), sham (1.06±0.35)] After been given drugs for30days, the AchE mRNA levels in Berberine-pinellia formula group was significantly drop, there was a significant difference (P<0.01). While Metoprolol group with not significantly difference [MI (4.58±1.33), metoprolol (3.79±1.58), large doses (1.44 ±0.72), and traditional dose (1.55±0.86)].â‘¡The TH mRNA levels in model group compared to the sham group was significantly increased, there was a significant difference (P<0.01)[MI (9.30±2.40), Sham (1.18±0.76)].After been given drugs for30days, the TH mRNA levels in Berberine-pinellia formula group was significantly drop, there was a significant difference (P<0.01). While Metoprolol group also came up with significantly difference [MI (9.30±2.40), metoprolol (1.90±0.60), large doses (2.06±0.89), and traditional dose (2.62±0.87)].Conclusion:Latest studies have shown that acute myocardial infarction(in30days) always accompany with multiple arrhythmia, cardiac autonomic neural remodeling is one of the key mechanisms of the arrhythmia. As the activity of Tyrosine hydroxyls and acetyl cholinesterase play an important role in evaluating the autonomic nerve. We can explore the effect of Berberine-pinellia formula in the acute phase of myocardial infarction by detect the activity of Tyrosine hydroxylsã€acetyl cholinesterase in myocardium. |
