Background and objective: Tic disorder is a common childhood-onsetneurobehavioral disorder,and nearly half of patients have one or morecomorbidies, which could affect children’s learning, life, social activities.It is anurgent need to be treated. Tiapride, as the first-line drug for the treatment of ticdisorder, is curative effect affirmation. Clonidine transdermal patch, which is forexternal use, is the new drugs for the treatment of tic disorder. It is simply andconvenient for changing once a week.To explicit the efficacy, tolerability andadverse reactions of monotherapy of Clonidine transdermal patch, a prospectivestudy is conducted.Method:160tic disorder patients, who were initial diagnosed or everdiagnosed and untreated, are selected from Jan2013to Dec2014. Inclusioncriteria: age from6y to18y; weight more than20kilogram; sex unlimited;corresponding to diagnostic standard of DSM-Ⅳ; never treated; Informedconsent of subjects (age>7y) and theheadofafamily. Dividing them into2groupsrandomly,we give clonidine transdermal patch for external use and tiapride fororal respectively. On the basis of YGTSS score untreated, compare YGTSSdeduction rate, treatment efficacy and adverse reactions through telephonefollow-up or face to face. Conduct blood and urine routine, liver and kidney function, blood glucose, blood lipid and electrocardiogram examination whenuntreated,four weeks and twelve weeks.Results:(1) Comparison of curative effects: after taking medicine for fourweeks, eight weeks and twelve weeks, excellent rates of clonidine transdermalpatch group and tiapride group are12.2%and20%,39.4%and42.1%,42.9%and44.6%respectively, and cure rates are0%and0%,19.7%and22.4%,32.9%and36.5%respectively. There is no difference between effectiveness of returnvisiting time among different groups.(2) Comparison of YGTSS deductionrate:There is no statistical significance between Motor twitch, voice twitch,functional damage of integral and total integral points deducted rate amongdifferent groups, after two weeks,four weeks, eight weeks and twelve weekstreatment.(3) Adverse reaction and retention rate:12cases suffered adversereaction in clonidine transdermal patch group (15.8%), and a total of21cases ofadverse reaction in tiapride group (25%). Retention rates of clonidinetransdermal patch group are100%、97.37%、93.42%and92.11%respectively,while those of tiapride group are98.81%、95.24%、90.48%and88.10%respectively. Although the retention rates of tiapride group are a little lower thanthose of clonidine transdermal patch group,but there is no statistical significancebetween the comparison of the two groups after12weeks treatment.Conclusions: Curative effect of clonidine transdermal patch, as first-episode tic disorder patients,is similar to that of tiapride. In addition, clonidinetransdermal patch have more wild slight adverse influence. Therefore,clonidine transdermal patch can be used as a conventional drugs for the treatment of ticdisorder.
