Research background:Ovarian cancer is one of the three big malignant tumor in female reproductivesystem, its incidence is second only to cervical cancer and endometrial cancer, but themortality rate of gynecological tumors of the first place, a serious threat to women’slife and health In patients with ovarian cancer to begin at home and abroad at presentUSES the standard treatment is the tumor cells to destroy the loss and platinum basedchemotherapy treatment, postoperative recurrence and metastasis rates are still high.Carboplatin (CBP) is the second generation of platinum anticancer drugs,,theadverse reactions such as renal toxicity neurotoxicity gastrointestinal reaction wereless than cisplatin, but studies have shown that carboplatin antitumor narrow range,and its side effects and drug resistance of limiting the clinical doses, so looking forrestrain and kill tumor cells and the effect of carboplatin chemotherapy drugs such asthe auxiliary drugs has become one hotspot of the ovarian cancer researchMelatonin(MLT) is secreted by the pineal gland a powerful indoles hormone,widely involved in human physiological activities, adjust biological rhythmsedative-hypnotic anti-stress antioxidant Strengthen the immune system, and so onCan also play them in a variety of ways to inhibit the action of the solid tumors, suchas breast cancer, prostate cancer bladder cancer pancreatic cancer melatonin asauxiliary chemotherapy drugs, and the joint application of carboplatin will enhancedestruction of ovarian cancer cells The role of inhibiting tumor cell apoptosis, reducethe side effects of carboplatin, and provides a new way for ovarian cancer treatment.Objective:To investigate the effect of melatonin combined with carboplatin on proliferationand apoptosis of ovarian cancer SKOV3cells in vitroMethods:(1) Human ovarian cancer SKOV3cells were cultured in vitro, the effect of separate and combined application of melatonin and carboplatin in ovarian cancerSKOV3cells was determined by MTT method.(2) The effect of separate and combined application of melatonin andcarboplatin in ovarian cancer SKOV3cells was detected by flow cytometry assay.(3) The effect of separate and combined application of melatonin and carboplatinin ovarian cancer SKOV3cells, Bcl-2、Bax expression were analyzed by RT-PCRassay.Results:(1) Melatonin carboplatin inhibite the proliferation of ovarin cancer SKOV3cells in a concentration dependentmanner, compare with carboplatin alone group, the combind application ofmelatonin carboplatin group significantly inhibited the proliferation of SKOV3cells,was statistically different(P<0.05)(2) Melatonin carboplatin inhibite the growth and induce apoptosis ofovarin cancer SKOV3cells in a concentration dependent manner, compare withcarboplatin alone group,the combind application of melatonin carboplatin groupsignificantly induce apoptosis of ovarin cancer SKOV3cells,was statistically different(P<0.05)(3) Melatonin carboplatin could increase he expressions of Bax andreduce the expressions of Bcl-2,compare with carboplatin alone group, thecombind application of melatonin carboplatin group significantly increase heexpressions of Bax and reduce the expressions of Bcl-2, was statistically different(P<0.05)(4) Melatonin plus carboplatin compared with application of carboplatinalone, reducing the amount of carboplatin, at the same time to achieve the same roleof value-added and promote apoptosis inhibition.Conclusion:(1) Melatonin carboplatin can inhibite the proliferation of ovarin cancerSKOV3cells and induce apoptosis of ovarin cancer SKOV3cells alone, but thejoint appli cation of inhibition rate and apoptosis rate significantly higher than thesingle drug group. (2) Melatonin carboplatin alone can increase SKOV3cells Bax gene expression,reduce the Bcl-2gene expression, joint application can obviously increase theexpression of Bax gene, significantly reduced the Bcl-2gene expression.(3) Melatonin plus carboplatin compared with application of carboplatin alone,reducing the amount of carboplatin, at the same time to achieve the same role ofvalue-added and promote apoptosis inhibition.
