Objective:The purpose of this study is to establish Carboplatin-resistance cell line, investigate the drug resistance mechanism and expression of indoleamine 2,3-dioxygenase of drug resistance ovarian cell lines SKOV3.Discussion was made about its relationship of drug resistance and IDOr. Study on the mechanism of resistant ovarian cancer metastasis or recurrence from the perspective of immunology by carboplatin induced multidrug resistance in ovarian carcinoma cell line effect on cellular immune function.Methods:The Carboplatin- resistance cell line,SKOV3/CBP,was induced by intermittent effect of medium dose Carboplatin;The proliferation of SKOV3 and SKOV3/CBP was tested by MTT; The invasion ability of SKOV3 and SKOV3/CBP was tested by Matrigel Plug;the level of indoleamine 2,3-dioxygenase was measured by immunoassay KIT.Then the two groups of cells were cultured with lymphocytes, C8+T cells separately, the killing ability of lymphocyte were tested by LDH release assay, the function of C8+T cells were detected by ELISPOT assay.Results:Compared with SKOV3 cell lines, the expression of IDO was significantly increased in SKOV3/CBP cell lines(P < 0.05)。Compared with the SKOV3 cell line, the proliferation ability of the drug resistant cell line was decreased, but the invasion ability was significantly enhanced. the killing capacity of lymphocyte, and CD8+T cell killingability of SKOV3/CBP cells group were significantly decreased(P < 0.05) than that of SKOV3 cells group.Conclusion:The metastasis of carboplatin resistant ovarian was occurred by cell invasion,rather than cell proliferation; SKOV3/CBP cells that Increased expression of IDO can reduce the sensitivity of the immune cells and immune escape, promote metastasis and relapse of chemotherapy resistant tumors further. IDO can be new promising targets for treatment of carboplatin resistance of ovarian carcinoma.
