Acute liver injury in acute clinical case is a serious threat to human health andpresent a rising trend year by year. Acute liver injury is a common liver disease. If nottimely treatment, it can lead to liver itself in synthesis, detoxification, excretion andbioconversion of severe metabolic disorders, eventually the body appear to clottingmechanism, jaundice, hepatic encephalopathy, and ascites as the main performance ofthe clinical symptoms. In recent years, with the rapid development of medicaltechnology, a variety of human diseases are the effective treatment. Acute liverdamage, however, the nature of the treatment of the disease status has not beenimproved.So to develop a new approach for the treatment of acute liver damage, is ofgreat significance in clinical application.Stem cell technology is one of the frontier technology in the field of regenerativemedicine research, in recent years, stem cell treatment also provides a new researchstrategy for a variety of diseases. This kind of stem cell therapy strategies for its sideeffects, efficacy significantly factors such as small, is widely recognized byresearchers, the treatment effect is significantly superior to the traditional mode ofdrug treatment.Especially bone marrow mesenchymal stem cells (BMSCs), due to itsmaneuverability, materials, training and so on in the treatment of various diseases hasa certain clinical application, in the treatment of acute liver injury, BMSCs also has acertain research foundation.In the process of acute liver injury, oxygen free radical on liver cell DNA structureand other metabolic process will have great influence, becoming the important causeof acute liver damage.As a member of the family of vitamins, vitamin C has thestrongest antioxidant effect, on oxygen free radical damage also had good resistanceeffect, the biological characteristics, make this kind of small molecular compound hasthe relief of acute liver damage course of the disease, liver damage, which has theapplication potential of the treatment of acute liver damage.This experimental study, which is used bone marrow mesenchymal stem cells andvitamin C to treat acute liver injury in rats in vivo and in vitro experiments respectively to observe the treatment effect, in the treatment of liver injury has someexploratory, and both the therapeutic effect of combined use of present moresignificant.Experiment which is used carbon tetrachloride injury were acute liver injury modelin rats.With bone marrow mesenchymal stem cells between combined effects ofvitamin C in rats with acute liver damage, on the level of rats in vivo and in vitro wereobserved on the improvement of the degree of acute liver injury.In vivo experiment,BMSCs (2.5x106cells/kg) and vitamin C (200mg/kg) in treatment of improvingliver function and liver index, the best treatment effect.On pathological change, thecombination group of liver tissue necrosis, ballooning in liver cells, fat degeneration,effectively relieve and inflammatory cells infiltration, etc.The immunohistochemicaldetection of liver tumor necrosis factor (TNF alpha, liver inflammatory index),bromine deoxyuridine (BrdU, bone marrow mesenchymal stem cell marker) showedthat between combined treatment group within the liver tissue of bone marrowmesenchymal stem cell number was significantly higher in treatment group alone, andthe expression of TNF alpha combined treatment group was obviously drop, thatvitamin C can promote bone marrow mesenchymal stem cells into hepatic injurybetween migration and enhance bone marrow anti inflammatory effect ofmesenchymal stem cells.Western blot-the results showed that the treatment group inthe liver Caspase3apoptosis gene expression have been cut, and the joint treatmentgroup effect is most obvious.Between vitamin C can promote bone marrowmesenchymal stem cells for the damage to the inhibition of cell apoptosis in theliver.In vitro experiments, vitamin C plays an promoting role in bone marrowmesenchymal stem cells between value-added, and with increasing concentration ofvitamin C have been promoting effect increased.Vitamin C is illustrated by the resultsof the optimization of mesenchymal stem cells in bone marrow.To sum up, BMSCs combined vitamin C in the treatment of acute liver injury inrats has better therapeutic effect.Vitamin C in the bone marrow mesenchymal stemcells to treat acute liver injury in rats can play in the process of optimization.So, thistopic not only developed a new biological treatment for acute liver damage model, and based on Vc in the thermal efficiency and easy to obtain, low toxic side effects ofBMSCs, the model also has a great clinical application value.
