PPAPs(polycyclic polyprenylated acylphloroglucinols)have good activities against tumor, virus and microorganism. The Path of its biosynthesis reveals that it’s easy for us to get new skeletons or new compounds. Many PPAPs were isolated from the plants of Hypericum species in the literature, such as Hypericum sampsonii and few reports concerning the chemical constituents of Hypericum sampsonii have appeared in the literature so far. At the same time, the chemical constituents of the same plant from different sources are different. So the aerial parts of Hypericum sampsonii which were collected from Da-bie Mountains in Qichun County, Hubei province were extracted and isolated in my study to get new compounds with changeable structures and have intriguing biological activities.From the extract,20polycyclic polyprenylated acylphloroglucinols were isolated by means of combination of silica gel, reverse silica gel, Sephadex LH-20, and semi preparative HPLC including two new compounds peroxysampsone C (1) and peroxysampsone D (2), seven ones were isolated firstly in this plant including otogirinin A (3), otogirinin B (4), otogirinin C (5), otogirinin D (6),28,29-epoxyplukenetione A (18), hypercohone A (19), and furohyperforin (20), and fifteen known compounds otogirinin A (3), otogirinin B (4), otogirinin C (5), otogirinin D (6), peroxysampsone A (7), peroxysampsone B (8), plukenetione C (9), plukenetione A (10), sampsonione Q (11), sampsonione J (12), sampsonione M (13), sampsonione N (14), sampsonione I (15), sampsonione C (16), sampsonione E (17). The structures of the compound were elucidated by NMR data and comparing with the date from the literatre.In addition, the cytotoxic activity against five human cancer cell lines of compound2was evaluated using MTS method, including HL-60(human promyelocytic leukemia cells), SMMC-7721(human hepatoma cells), A-549(human lung adenocarcinoma cells), SK-BR-3(breast cancer cells) and SW480(human colon cancer cells). Compound2showed moderate cytotoxic activity against five human cancer cell lines with IC50values ranging from3.83-19.63μM.
