PART1Protective effects of shengmai san on H9c2Cardiomyocytes exposed to highglucoseObjective: To explore protective effects of shengmai san on H9c2Cardiomyocytesexposed to high glucose.Methods: H9c2Cardiomyocytes were randomly divided into two groups: extracts groupand monomer recipe group. We screened optimum concentration of normal H9c2cellsthrough MTT assay,and judged the protective effect of shengmai san on H9c2Cardiomyocytes exposed to high glucose by cellular survival rate.Results: The study displayed the normal H9c2cells had the highest survival rate in theconcentration of100μL. The survival rate of the concentration of100μL was higherthan other concentrations (P <0.05). The survival rate of both groups exposed to highglucose raised with increasing concentration. The concentration of100μL reached thehighest survival rate, with a significant difference with the other groups (P <0.01).Conclusion: Protective effects of extracts and monomer recipe of shengmai san on H9c2Cardiomyocytes exposed to high glucose. PART2Study on the effect and mechanisms of shengmai san effective componentsagainst myocardial apoptosis induced by high glucose.Objective: To explore mechanisms of shengmai san effective components againstmyocardial apoptosis induced by high glucose.Methods: Primitive cardiomyocytes of wistar neonate rats were randomly divided intoeight groups: control group, model group, extracts groups with various concentrations(100μL,50μL,10μL shengmai san extracts-containing serum) and monomer recipegroups with various concentrations (100μL,50μL,10μL shengmai san monomer recipe-containing serum). In addition to the control group (concentration ofglucose:5.5mmol/L), other groups have maintained a high concentration of glucose(25mmol/L). Flow cytometry was designed to observe and detect cellular apoptosis;western blotting analyzed expression of p53,Bcl-2and Bax; Q-PCR detected transcriptlevels of Fas and FasL.Results: Compared with the control group, apoptosis was significantly increased inModel group (P<0.01). The apoptosis rate of both extracts groups and monomer recipegroups decreased with increasing concentration. Compared with model group, apoptosiswas significantly decreased in other groups (P<0.01). Compared with model group, thelevel of Bcl-2decreased and the level of Bax and p53increased significantly in bothextracts groups and monomer recipe groups (P<0.01). Compared with model group, thetranscript level of FASL decreased and the transcript level of FAS increasedsignificantly in control group (P<0.05). Other groups have no difference with modelgroup.Conclusion:Inhibitory effects of shengmai san effective components on Cardiomyocytesapoptosis induced by high glucose. The mechanisms of inhibitory effects may associatewith blocking mitochondrial apoptosis pathway. And the research showed similareffects between extracts groups and monomer recipe groups.
