To investigate the expression of COX-2and P53in breast premalignant lesions ofhunman and to research Yanghehuayan-decoction’s inhibition effect on rats with breastpremalignant lesion and the expression of COX-2and P53.To explore the interventioneffect of Chinese medicine on breast premalignant lesions.Methods:(1)Choose18patients from cyclomastopathy and investigate the expressionof COX-2and P53in breast tissue with immunohistochemistry. So do the precancerosisand breast carcinoma groups.(2)Molding and grouping of zoopery:To induce breastpremalignant lesion model of mouse with DMBA except the blank group.After successfulmodeling grouping the mouse into Yanghehuayan-Decoction group,Sanjie-decoction groupand disease model group,Pingxiao-tablet group and tamoxifen group,and the treatmentgroups were given corresponding drugs.Observing pathological morphology of breasttissue by HE staining,detecting the cox-2and p53expression positive rate and gray valueby immunohistochemistry.Results:(1) Clinical studies showed:The positive expression of cox-2in breastpremalignant lesions was:49.8%±3.46, the difference between breast premalignant lesionsand breast cancer showed no statistically significance (P>0.05); the difference betweenbreast premalignant lesions and breast hyperplasia had statistically significance (P<0.05).The positive expression of P53in breast premalignant lesions was:45.45%±5.25,there were obvious differences between groups(P<0.01).(2) On the incidence of rat breast premalignant,all treated groups showed significantdifference compared with the disease-model group(P<0.05). As Yanghehuayan-decoctiongroup, the incidence rate of breast premalignant lesion and invasive breast cancer was47.06%and29.41%, which showed no significant difference compared with other treatedgroups(P>0.05).(3) Compared with the disease model group, other groups,positive rate and gray valueof cox-2and p53have significant difference between each other (P<0.05).The effect ofcox-2of Yanghehuayan decoction group was45.71%±3.96,which showed no significantdifference compared with Sanjie decoction group(P>0.05),the rest groups have significantdifferences between each other.The positive expression rate of P53of Yanghehuayandecoction group was41.18%±3.45,compared with Sanjie decoction group,tamoxifengroup and Pingxiao-tablet group,the difference was statistically significant(p<0.05).Conclusion:(1) The positive expression of COX-2and P53showed a rising trend withthe evolutionary process of breast hyperplasia,precancerous lesion to invasive cancer.(2)Wenyangsanjie prescription-Yanghehuayan decoction could blocking or delayingthe progression of breast cancer,and inhibiting the expression of COX-2and P53to acertain extent.
