Objective: Observing the expression of lymphatic factor VEGF-C and its receptorVEGFR-3in human breast cancer tissues of precancerous lesions.Researching the effect ofYanghehuayan-decoction on breast precancerous lesions of VEGF-C and VEGFR-3,so asto explore the mechanism of traditional Chinese medicine for treating breast precancerouslesions.Methods: Clinical research by selecting20cases of patients with breast glands, breastprecancerous lesion and breast cancer.Using Immunohistochemical method detects theexpressions of VEGF-C and VEGFR-3in various pathological types in organization.Ratmodels of breast premalignant lesion were established by DMBA.After9weeks,they weregrouped randomly as disease group,the group of Yanghehuayan-decoction, the group ofSanjie-decoction,the group of Pingxiao-tablet,and tamoxifen group.All were treated bywater,Yanghehuayan-decoction(7.2g/kg/d),sanjie-decoction(5.4g/kg/d),Pingxiao-tablets(144mg/kg/d)tamoxifen(4mg/kg/d)separatedlyfor4weeks.Observing pathological morphologyof breast tissue by HE staining,the experiment immunohistochemically method to detectthe expressions of VEGF-C and VEGFR-3. By means of SPSS17.0data software package,it begin to dispose that by means of statistics.Results:(1) the clinical research shows that the positive expression rates of VEGF-Cin hyperplasia of breast glands, breast precancerous lesion and breast cancer arerespectively14.25±2.63、19.5±10.93、29.5±19.03.There is a clear difference in theexpressions(p<0.05). And that of VEGFR-3are respectively4.50±2.04、12.07±2.47、14.45±3.88(p<0.01). The researching result of animals: the incidence of rat breast premalignant, positive and intensity of VEGF-C, VEGFR-3(except Pingxiao-tablets)alltreated groups showed significant difference compared with the disease group(p<0.05).As Yanghehuayan-decoction group, the incidence rate of breast premalignant lesionand invasive breast cancinoma was47.06%and29.41%, which showed no significantdifference compared with other treated groups(p>0.05).The gray value and positive rateof VEGF-C are135.60±8.23、20.98±5.37,which are compared with the other treatmentgroups, besides tamoxifen group,the differences were statically significant(p<0.05). Thegray value and positive rate of VEGFR-3are146.59±12.79、13.06±2.27, which arecompared with the other treatment groups, besides tamoxifen group, the differences werestatically significant(p<0.05).Summary: Breast glands organization of human body,the expression capacities ofVEGF-C and VEGR-3are the weakest in hyperplasia of breast glands,but relatively strongin breast precancerous lesion and the strongest in breast cancer. The decoction ofYanghehuayan,tamoxifen,Sanjie-decoction,Pingxiao-tablets intragastrically breast tissue ofrats with malignant disease group is significantly lower than that in. Breast tissue ofnormal rats VEGF-C, VEGFR-3showed low or no expression in breast tissue, bytamoxifen, and Yanghehuayan-decoction, Sanjie-decoction, Pingxiao-tablets orally in ratsVEGF-C, VEGFR-3expression gradually decreased;Because the Pingxiao-tablets has nodifference with disease group,so the Yanghehuayan-decoction, Sanjie-decoction, amine ofthree benzene oxygen intervention breast precancerous lesions maybe have a certain role.Expression and the Yanghehuayan-Decoction group in the tamoxifen group, lower than theother treatment groups, and the effect was better than that of Sanjie-Decoction andPingxiao-tablets, which had no significant difference as compared with tamoxifen, twowere decreased the expression of regulatory factors, further affirmed and the decoction ofYanghehuayan by reducing the feasibility to inhibit lymphatic generation rate of positiveexpression of promoting lymphangiogenesis factors.
