OBJECTIVE Study on the pilot process of total saponins in Lysimachia capillipesHemsl.to make improvements to increase the content of total saponins. and thechitosan flocculation process of the total saponins intermediate solution of Lysimachiacapillipes Hemsl. and lay the foundation of medium pressure column separator thetotal saponins monomer components. METHODS By analysis of saponin content,the extraction and purification pilot process were optimized and the quality standardsof intermediate was also developed. By Visible spectrophotometry and HPLC-ELSD,flocculation rate and the total saponins loss rate were used as two main parameters toestimate the effect of influencing factors on flocculation. The optimum conditions ofChitosan flocculation was determined by orthogonal test. RESULTS The saponincontent and yield were60%and3%in the optimized pilot process. The optimumconditions: Each100ml intermediate solution,aqueous solution concentration of0.6mg/mL, the dosage of chitosan solution30ml, flocculation temperature60℃,flocculation time4h, and pH4.12. Under these conditions, a flocculation rate of47.81%and a total saponins loss rate of9.53%were achieved.
