| Background:Surgery and chemotherapy become basic therapy for GC in the worldwide, the publication of studies have shown chemotherapyimproved survival, however,there is no standard chemotherapy protocols for advanced gastric cancer. Fluorouracil combined with platinum as common chemotherapy strategy is the most efficient therapy in advanced gastric cancer Chemotherapy curative effect and adverse events exist the differences between individuals in clinical. At present, the publication of studies showed polymorphism of MTHFR C677T gene may be associated with sensitivity to fluorouracil-based chemotherapy in AGC, but there is a matter of controversies. Therefore, our meta-analysis may provide fundamental research, Multi-Disciplinary Treatment and individualized treatment with evidences. Purpose:To evaluate the relationship between C677T Polymorphysim of MTHFR gene and efficacy to fluorouracil-based chemotherapy in advanced gastric cancer (AGC). Methods:The published case-control studies on C677T gene Polymorphysim of MTHFR and Sensiticity to Chemotherapy in AGC were obtained from China Journal Full-text Database, Chinese Science and Technology Periodicals, Wan Fang databases, Chinese Biomedical Literature Database, PubMed, EMBASE, Cochrane library and Web of science with computer. RevMan5.1software was used to conduc the statistical analysis. Results:A total of775cases from7case-control trials were included in the meta analysis.The result of meta analysis showed amang MTHFRC677T, The OR and95%confidence interval (CI)of TT vs CC was4.63(1.23-17.4), CC+CT vs TT was0.21(0.06-0.78), Subgroup analysis showed the OR and95%CI of TT vs. CC was32.99(11.40-95.42), CC+CT vs TT was0.04(0.02-0.10) in Asians, statistical significance was found. After the Sensiticity analysis based on different detection methods, the results showed there was a significant difference in TT vs CC and CC+CT vs TT, the OR and95%CI was6.03(1.53-23.72) and0.17(0.04-0.68) Respectively. Conclusion:Polymorphism of MTHFR C677T gene may be associated with sensitivity to fluorouracil-based chemotherapy in AGC. |
