Objective Observed in different periods after corneal alkali burns of histopathologicalchanges, the expression of leptin and pigment epithelium derived factor, as well asacetylcysteine impact on leptin and pigment epithelium derived factor, explore their relationshipwith corneal neovascularization.Methods CNV models were established by putting the filter paper with the diameter of3mm erased into1mol.L-1NAOH solution on the central corneas of right eyes of rats for30seconds. Right eyes of75vista rats were treated as experimental group and divided into3groups A、B and C,25eyes in each group. Left eyes were treated as normal control group.Starting from the date of the burn group a drip PBS; Group B drip acetylcysteine5times a day;Group C within7days after burns drip PBS, been added drop of acetylcysteine5times a dayfrom7days to14days. We use slit lamp microscope to observe CNV every day and calculatelength and area of CNV.Then detected it by HE staining and mmunohistochemical method.Results B group corneal neovascularization growth rate is lower than that of A、C afteralkali burn for4days and7days; The area measured of corneal neovascularization is that B <C<A. Immunohisto chemical detection showed that1d after corneal alkali burn, the expressionof leptin and pedf begins to rise. The expression of leptin at7thday after injury: A(0.2761±0.1144);B(0.2322±0.0749);C(0.2692±0.0646).The expression of leptin overall havedifferences (F=159.423,P <0.05).The differences between A and B、B and C were statisticallysignificant after the multiple comparison (P <0.05), expression of leptin in group B is lowerthan in group A and group C. There was no statistically significant difference although the meanof group A is greater than group C(P>0.05).The expression of pedf: A(0.3521±0.1074),B(0.3899±0.0721); C(0.3561±0.1233). The expression of pedf overall have differences(F=418.947,P <0.05). The differences between A and B、B and C were statistically significantafter the multiple comparison (P <0.05), expression of pedf in group B is lower than in group Aand group C. There was no statistically significant difference although the mean of group A islower than group C (P>0.05).The expression of leptin at14thday after injury: A(0.3161±0.0803);B(0.2504±0.1279);C(0.2823±0.0776).The expression of leptin overallhave differences (F=651.837,P <0.05), the differences between every groups were statisticallysignificant after the multiple comparison(P <0.05),B <C <A.The expression of pedf at14thdayafter injury: A(0.2234±0.1186),B(0.2530±0.1013), C(0.2377±0.1067).The expressionof pedf overall have differences (F=735.288,P <0.05), the differences between every groupswere statistically significant after the multiple comparison(P <0.05),B>C>A.Conclusion: Expression levels of leptin and pedf have relationship with CNV.Acetylcysteine can effectively reduce the expression of leptin, enhance the expression of pedf.
