Object: By means of investigating the levels of serum interleukin-2,interferon-γ, interleukin-4and interleukin-10in these patients with unexplained recurrent spontaneous abortion, normalpregnancy women and normal non-pregnant women in gestation age, we are able to realizethe different levels of TH1/TH2cytokines in unexplained recurrent spontaneous abortionpatients for exploring the mechanism of spontaneous abortion further, to provide the latestimmunized theory for diagnosing and curing these patients with unexplained recurrentspontaneous abortion in the clinical and afford pertinence case selection theory for promotingactive immuntherapy preferably in the future.Methods:40patients with unexplained recurrent spontaneous abortion in the obstetricdepartment of our hospital were collected as the case group,40normal early pregnancywomen as one matched group and40non-pregnant women in gestation age as anothermatched group from October2012to October2013.Collecting8ml fasting peripheral venousblood, spare frozen the supernatant at-80℃after centrifugation. ELISA was used to quantifythe levels of TH1cytokines(IL-2、IFN-γ) and TH2cytokines(IL-4、IL-10) of the three groups,analyze the otherness of TH1/TH2cytokines between case group and matched groups; analyzeotherness of change value of TH1/TH2cytokines between case group and matched groups byusing the average levels of non-pregnant women’s cytokines as the base value; and analyzethe correlation of TH1/TH2cytokines in three groups.Results:(1)TH1cytokines: the levels of IL-2and IFN-γ in case group are apparently higherthan matched groups with a conspicuous otherness (P<0.01),but there was no significantdifference between the two matched groups(P>0.05).(2)TH2cytokines: the levels of IL-4andIL-10in case group are apparently lower than matched groups with a conspicuous otherness(P<0.01),but there was no significant difference between the two matchedgroups(P>0.05).(3)The change values of IL-2and IFN-γ in URSA group are higher than thepregnancy group and there was a significant difference(P<0.05);the expression of IL-4andIL-10shows a decreasing trend in the case group, the change value of IL-4and IL-10areapparently higher than the pregnancy group with a conspicuous otherness (P<0.05);(4) Therewas positive correlation between IFN-γ and IL-4in non-pregnantwomen(r=0.368,P=0.02,P<0.05) by the Pearson correlation analysis in TH1/TH2cytokines ofthese three groups, but other data correlation analysis can not yet indicate a correlationbetween TH1/TH2type cytokines.Conclusion:(1)The levels of TH1cytokines(IL-2and IFN-γ) in case group are apparentlyhigher than the matched groups that explain the high expression of TH1cytokines in maternalperipheral blood is not conducive to the normal pregnancy.(2)the levels of TH2cytokines(IL-4and IL-10) in case group are significantly lower than two control groups that explain normal pregnancy is a kind of "TH2phenomenon", and the low expression of TH2cytokines inmaternal peripheral blood will result in spontaneous abortion.(3)There was no statisticallysignificant difference in the levels of TH1/TH2cytokines between the two matchedgroups.(4)By using the average levels of non-pregnant women’s cytokines as a base value, theTH1cytokines expresses positively and TH2cytokines expresses negatively, the othernessbetween them was conspicuous(P<0.05),that explain that higher express of TH1cytokines isnot conducive to normal pregnancy, and immune balance between matrix and embryo is animportant condition for normal pregnancy, the imbalance of maternal immune system willlead to the occurrence of spontaneous abortion.(5)There was positive correlation betweenIFN-γ and IL-4in non-pregnant women(r=0.368,P=0.02,P<0.05) through the Pearsoncorrelation analysis in TH1/TH2cytokines of these three groups, but other data correlationanalysis can not yet indicate a correlation between TH1/TH2cytokines.
