Objective:The RhoBTB family is a member of the RhoGTP family. Unlike the functionof other typical Rho family members in the regulation of cell proliferation, adhesion,migration and membrane transport, RhoBTB subfamily’s function is not clear. Somestudies have confirmed that RhoBTB protein in tumorigenic process which is caused byprotein dysfunction and alterations in cellular homeostasis. RhoBTB research is notonly beneficial to the further understanding of the Rho family, and is more helpful toreveal the pathogenesis of tumors and tumor targeting drug research and development.This paper from the molecular, cellular, and clinical aspects studied the biologicaleffects of RhoBTB3in renal clear cell carcinoma.Methods: To detect the expression of RhoBTB3in renal cell carcinoma and theadjacent normal tissues, and renal clear cell carcinoma cell line and normal renal tubularepithelial cell line By RT-PCR, immunohistochemistry and Western blot method. Toconstruct the eukaryotic expression lentiviral vector of pLVX-IRES-Neo-RhoBTB3andpLVX-IRES-ZsGreen-RhoBTB3, and infect786-o and HKC cell lines. MTS methodwas used to detect the characters of RhoBTB3in the regulation of ccRCC cellproliferation.Flow cytometry was used to detect the effect of RhoBTB3on renal cancercell cycle and apoptosis.Result: RT-PCR, immunohistochemistry and Western blotting showed that theexpression of RhoBTB3in renal clear cell carcinoma tissues and cell lines wassignificantly lower than that in the corresponding adjacent tissues of carcinoma andnormal renal tubular epithelial cell line (HKC)(P <0.05). The overexpression ofRhoBTB3significantly promotes the apoptosis of786-O cells in renal cell carcinoma,and Silencing RhoBTB3induces HKC cells from G1to S phase, which promotes cellproliferation, and promotes the invasion and migration of HKC cells.Conclusion: The expression of RhoBTB3in clear cell renal cell carcinoma tissues and cell lines is lower than that in the corresponding adjacent tissues of carcinoma andnormal renal tubular epithelial cell line. Overexpression of RhoBTB3promotes cellapoptosis and inhibits cell proliferation. Down-regulation of RhoBTB3induces cellcycle G1/S transition and promotes cell proliferation. Inhibition of the malignantprogression of RhoBTB3in clear cell renal cell carcinoma can provide a new directionfor molecular target to treatment of renal cell carcinoma.
