Tanshinones are main lipophilic active components in Salvia miltiorrhiza Bunge which could anti-bacterium, anti-inflammation and anti-knub. In the paper, the tanshinones extraction technics and preparation technics of tanshinones’ industrial dynamic axial chromatography monomer were studied.Based on the evaluating index of contents of salivianolic acid B, danshensu, tanshinone ⅡA and crypototashinone which were determined by HPLC and TLC, the effects of ten kinds of solvents on extraction and extraction order of salivianolic acid B, danshensu, tanshinone ⅡA and crypototashinone from salviae miltiorrhizae bunge were systematically investigated. The results showed that the beneficial order of extraction was fat-soluble components priority and then water-soluble components been extracted. The dichloromethane was suitable for the extraction of fat-soluble components. The methods of comprehensive extraction of active components from salviae miltiorrhizae bunge were established. Fat-soluble compounds were extracted with dichloromethane first and then water-soluble compounds were extracted with methanol.The fat-soluble tanshinones were firstly separated by industrial dynamic axial chromatography with dichloromethane and petroleum as mobile phase in gradient elution. The solid phase of the chromatography was silica and the column chromatography conditions have been determined by TLC.100kg root part of salvia miltiorrhiza bunge was extracted with dichloromethane and separated by industrial dynamic axial chromatography three times. Finally, four kinds of tanshinones such as141.5g tanshinone Ⅱ A,12.5g tanshinone Ⅰ,73.2g cryptotanshin and8.2g unidentified compound BC-3were obtained. The purity of four compounds were98.5%,96.2%,97.8%, and95.4%respectively. The structures of BC-3need identifying more by another methods, such as HNMR,13CNMR. The chromatography run time was less than8h per column.In conclusion, the industrial dynamic axial chromatography have many advantages, such as simple operation, short separation time, high efficiency, which have an important significance to separation of natural active components.
