Polysaccharides were extracted and purified from abalone gonad and scallop viscera, and their antitumor effects were investigated.Crude polysaccharides from abalone gonad and scallop viscera were extracted by enzyme hydrolysis, named as CAGP and CSVP, respectively. They were purified by Sephadex G-100gel chromatography, and AGP-I and SVP-I were thus obtained correspondingly. Component analysis showed that the polysaccharide contents of AGP-I and SVP-I were34.65%and48.73%, the protein contents were33.6%and25.1%, the sulfate contents were9.19%and5.14%, the aldonic acid contents were18.02%and14.89%, respectively.Antitumor activity in vitro showed that CAGP and AGP-I could restrain the proliferation of A549, Hela and K562cells. Their inhibition rates of800μg/mL dose were23.84%and33.79%to A549,26.53%and24.20%to Hela,13.77%and12.83%to K562. CSVP and SVP-I could also restrain the proliferation of A549, Hela and K562cells. Their inhibition rates of800μg/mL were27.43%and34.22%to A549,24.26%and19.62%to Hela,26.60%and26.73%to K562, respectively.Antitumor activity in vivo showed that AGP-I and AVP-I could restrain the growth of H22tumor. The inhibitory rate of CTX is69.21%,40.07-49.03%for AGP-I of50-200mg/kg dose,51.97-62.37%for AVP-I at50-200mg/kg dose. Moreover, AGP-I and AVP-I could markedly increase spleen weight, lymphocyte proliferation, phagocytosis of macrophage, natural killer cell activity, and prodution of cytokines, such as IL-1, IL-2, IL-4, TNF-a and IFN-y in tumor bearing mice. SVP-I could inhibit H22tumor growth, with an inhibition rate of33.74%at200mg/kg dose, and markedly increase the spleen weight of tumor bearing mice.The above results suggested that the polysaccharide from the abalone gonad, abalone viscera and scallop viscera had antitumor effect, which may act by increasing the immune function of tumor-bearing mice. These data provide a theoretical basis for fine processing and comprehensive utilization of abalone and scallop viscera.
