This article has systematically studied Poria Cocos harvest from geoherbsregions Yunnan (YN) and Dabieshan (DBS) by infrared spectroscopy,basedon the work in the natural resource survey of Poria Cocos,builded a set of InfraredSpectra method suitable for rapid discrimination of Poria Cocos from differentgeographical regions.1.The geoherbs producing area of Poria Cocos in YN and DBS were surveyedby the methods of consulting the literature review,asking the experts,accessing toseminars,field investigation and other methods,finished the research data ofecological environment,cultivation techniques,breeding strains,the distributionof output and sales price,visited to the large Chinese herbal medicine markets atthe same time.The result showed that the wild Poria Cocos were very rare throughthe changes of area.The standardized production standards of Poria Cocos shouldbe formulated as soon as possible to strengthen macro-control.Special attentionshould be paid to the protection,research,promotion of traditional,high-qualityvarieties of Poria Cocos.2.The traditional identification methods was used to discriminated thecharacter of poria geoherbs collecting in filed sample survey.The following traitswere considered much better were that a weight solid mass,brown skin color,striaefine with no cracks,white delicate cross-section and strong sticky teeth.Most offresh mature Poria Cocos harvested from YN were spherical,big and weight,butflat round or irregular lumps,small,light harvested from DBS.The processspecifications of Poria Cocos were roll,sheet,block in YN for the main purpose ofexport,but small block in DBS for the main needs of pharmaceutical factory.3. Infrared Spectroscopy(IR)combined with Chemometrics software wasused to identification and recognition of Poria Cocos from different geographicalregions.Though analyzing the IR spectra of the powder and raw materials of PoriaCocos,the IR characteristic absorption peaks of certain constituent of Poria Cocospolysaccharides are found:1163,1078,1035and995cm-1,it can be used fordiscrimination of Poria Cocos.Analyze the quality of different sources of PoriaCocos herbs though the IR technology.Significant difference appeared between inPoria cocos and its adulterants circulate on the market.Small significantdifferences appeared in the Poria Cocos polysaccharide among different geographical regions.There are differences in poria cocos polysaccharide on thecomponent content among the different cultivation way of Poria Cocos at the samegeographical regions.the IR characteristic absorption peaks of certain constituentof Poria cocos peel were1149,1079and1036cm-1(stretching vibration ofpolysaccharide C-O),1619,1315and780cm-(1IR characteristic absorption peaksof calcium oxalate),3696,3621,797,779,537and470cm-1(IR characteristicabsorption peaks of kaolin),it can be used for discrimination of Poria cocospeel.Clear differences in Poria cocos peel due to the large difference of cultivationenvironment and the results were consistent with Poria Cocos.The non-destructiveand rapid detection of IR,could objectively reflect the main component of Poriacocos,as well as overall composition in the macro.It suggested that IR couldpreliminary identify of Poria Cocos from different origin sources and analysiscomposition.4.Mid-Infrared Spectroscopy combined with the extraction process of Poriacocos.Though analyzing the IR spectra of the ethanol precipitate from the aqueousextracts of samples show that the IR characteristic absorption peaks of certainconstituent were1655cm-1(IR characteristic absorption peaks of amide Ⅰ bandof protein),1545cm-1(IR characteristic absorption peaks of amide Ⅱband ofprotein),1123and1080cm-1(stretching vibration of polysaccharide C-O),619cm-1(IR characteristic absorption peaks of potassium sulfate).The differences inthe peak area and intensities at protein and potassium sulfate as well as thecomposition of polysaccharide were appeared among these ethanolprecipitate.Although their corresponding residues obtained after water extractionexhibited only small differences observed from the IR spectra suggested that mostPoria cocos polysaccharide were water insoluble.For the first time analyzing theraw materials,the ethanol precipitate from the aqueous extracts and thecorresponding residues obtained after water extraction of Poria Cocos at the sametime,to provide new ideas for the overall analysis of Poria Cocos and traditionalChinese medicine....
