Background and Objective:In1983, helicobacter pylori was found and cultured by Marshall and Warren successfully. The correlation and mechanism between Hp and diverse of chronic diseases, precancerous diseases, gastric cancer were attracted by many scholars. The phenomenon that Hp induce gastric epithelium cell apoptosis and caused compensatory cell hyperplasia at the same time, so which maybe cause pathogenetic mechanism of gastric cancer. However, it’s unclear that Hp unbalance the system of apoptosis and hyperplasia. To investigate the effect of.Hp-induced alteration of cell proliferative activity and apoptosis on gastric epithelium and cancer cell from change of cell morphology, protein expression of proliferation,cell cycle and apoptosis.Methods:(1) The morphological changes were observed by inverted microscope after GES-1, SGC-7901, MGC-803and BGC-823treated by Hp for24h and48h.(2) MTT assay was used to test the cell proliferative activity in four kinds of cells.(3) Flow cytometry was used to detect the cell cycle and apoptosis of the cells.(4) The expression of Caspase-3,-9, P53, P21and E2F1was detected by western-blot. Results:(1) The morphological changes in four kinds of cells were observed24hours later after treating by Hp and the changes were more significant with the increase of Hp.(2) MTT showed that cell proliferative activity in four kinds of cells were inhibited by Hp.(3) FCM showed that cells in S phase increased but cells in G1phase decreased.(4) The expression of caspase-3,-9was decreased in four kinds of cells to different extent. The expression of P53, P21and E2F1was low-expressed or decreased expression.Conclusions:Hp can induce the abnormal change of cell morphology and proliferative activity in gastric epithelium and cancer cells, and Hp affect cell proliferation and apoptosis on four cells to different extent.
