Objective To observe the influence of Helicobacter pylori(Hp) NCTC11637 on growth of SGC-7901 cell by constructing cell acute infectious model in vitro, determinating cyclin dependent kinase inhibitor p27kipl,its mRNA and expression of adhesion molecule CD54,CD44s, in order to explore the function of cell cycle regulatory element and adhesion molecule in the development of gastric cancer, offer theory basis for clinic diagnosis ,prevention and cure of gastric cancer. Methods Construct the infectious model by adding NCTC11637 of six different kinds of concentration(3.8×104 ~1.2×108CFU/ml)to gastric cancer epithelium cells SGC-7901. Cell without adding Hp is served as control .Do the following experiments with cell infected by Hp of different kinds of concentration and cell of control . (1) analyze DNA ploid ,determinate cell apoptosis and calculate Proliferation Index(PrI)through flow cytometry(FCM), and also examinate antigen protein of adhesion molecule CD54,CD44s . (2) Examinate proliferation of cell nuclear antigen (PCNA) and p27kipl antigen by immunocytochemical analysis .(3) Determinate mRNA of p27kipl by cell situ hybridisation analysis.(4) Observe cell apoptosis by technology of transmission electron microscope(TEM) ,and Observe change in cell Morphology by technology of light microscope(LM) . Result (1)expression of PCNA antigen and PI was enhanced when cells of SGC-7901 coincubated with the Hp in low concentration(≤1.9×105CFU/ml) but inhibited in higher concentration(≥4.8×106 CFU/ml) .(2)FCM indicate :group control compared with groups 3.8×104CFU/ml and 1.9×105CFU/ml, cell apoptosis rate has no significant difference(P>0.05).While group of Hp concentration ≥9.6×105CFU/ml compared with group control ,cell apoptosis rate has been enhanced and has significance(P<0.01).(3)Groups of different concentration of Hp compared with group control the express of p27kipl has been Reduced significantly showing a dose-dependent picture, express of mRNA has neither significant differenc(eP>0.05)nor consistence with express of p27kipl. Intensity of average fluorescence determinated by FCM manifest that express of CD44s has been enhanced significantly while express of CD54 disappear in various degree showing a dose-dependent picture.(4)LM manifest : coincubated with different concentration of Hp mostly cells diminish them volume showing variety of shapes, Some cell nucleus contract and cracke, have a big nuclear or many nucleus, Some cells show vacuolar pathological change.(5)TEM manifest :epithelium cells infected by different concentration of Hp are all have vacuolar pathological changes and cell nuclear contraction, nuclear chromatin gathering around , which is early apoptosis. Conclusion Infection of Hp may destroy normal regulation of cell cycle, and also cause unbalance of cell apoptosis and cell proliferation, which has offered biology basis for gastric cancer, abnormal expression of adhesion molecule have accelerated the occurance and development of cancer. Result suggest :In different stage of infection Hp play an important role to development of gastric cancer by different mechanism, on which we will make further research.
