The technique of immunohistochemistry(IHC) and In situhybridization(ISH) were used to detect the expressions of SPARC, TGFβ1,Endoglin in lung cancer and normal lung tissues, thus to investigate thecorrelation between clinical pathological features and the expressionswhich SPARC, TGFβ1, Endoglin mRNA and their protein had in lungcancer and adjacent normal lung tissues; to investigate the relationshipsamong expression of SPARC, TGFβ1, Endoglin mRNA and their proteinin lung cancer and its relationship with angiogenesis.Collect54cases with lung cancer and2cases of adjacent normallung tissues by using tissue chip and prepare tissue chip by selecting, themicrovascular morphology of different vascular dense tissue microarray.By using Immunohistochemical staining respectively by CD34,Endoglinand α-SMA markers, observe the relationship between the microvesselwall cell immunophenotype and different morphological features, andselect the specific marker of neovascularization.The protein and mRNA expressions of SPARC, TGFβ1and Endoglinprotein, were detected in71cases of lung cancer tissues, and30casesof adjacent normal lung tissues through the technique ofimmunohistochemistry (IHC) and in situ hybridization (ISH) respectively.The correlation between the three indexes and such clinical andpathological features of patients as age, sex, the size of tumor, the degreeof tumor differentiation, clinical classification, TNM stages and lymphnode metastasis, was analyzed, the correlation among the three indexeswas analyzed, and the consistence of expression of SPARC, TGFβ1,Endoglin was detected between the two techniques of immunohistochemistry (IHC) and in situ hybridization (ISH).As the protein expressions of SPARC, TGFβ1, Endoglin wasdetected in71cases of the lung tissues and30cases of adjacent normallung tissues through the technique of IHC. The positive expression ratesof SPARC protein, TGFβ1protein were56.34%(40/71),60.56%(43/71)and mark Endoglin protein in lung cancer MVD average was12.45±6.53respectively in lung tissues, and apparently higher than20%(6/30),30%(9/30) and0.31±0.46in adjacent normal lung tissues, the variancewas significant (P<0.05); and the high expression of SPARC andEndoglin protein in lung tissues had correlation with the TNM stages andlymph node metastasis, however, it didn’t have relationship with theclinical and pathological features such as age, sex, tumor size,differentiate degree and clinical classification. The high expression ofTGFβ1protein in lung tissues had correlation with tumor size, TNMstages and lymph node metastasis, but didn’t have relationship withpatients’ age, sex, the degree of tumor differentiation and clinicalclassification. The expression of SPARC protein and TGFβ1protein waspositive signal. The expression of SPARC protein increased MVDdecreased gradually, However TGFβ1protein increased MVD increasedgradually, SPARC, TGFβ1were correlated with MVD(P<0.05).As the expressions of SPARC mRNA, TGFβ1mRNA was detectedin71cases of the lung tissues and30cases of adjacent normal lungtissues through the technique of ISH, the positive expression rates ofSPARC mRNA, TGFβ1mRNA were61.97%(44/71),59.15%(42/71) andapparently higher than20%(6/30),26.67%(8/30) in adjacent normal lungtissues, the variance was significant (P<0.05); and the high expression ofSPARC mRNA in lung tissues had correlation with the TNM stages andlymph node metastasis, however, it didn’t have relationship with theclinical and pathological features such as age, sex, tumor size,differentiate degree and clinical classification. The high expression of TGFβ1mRNA in lung tissues had correlation with tumor size, TNMstages and lymph node metastasis, but didn’t have relationship withpatients’ age, sex, the degree of tumor differentiation and clinicalclassification. The expression of SPARC mRNA and TGFβ1mRNA wassignificantly positive.Progress of high expression of SPARCmRNA inlung cancer may inhibit lung cancer progression, and high expression ofTGFβ1mRNA may promote lung cancer, and participate in the transferprocess of lung cancer, which may play an important role.Of71tissue specimens of lung cancer, the positive expression ofSPARC protein were40cases, SPARCmRNA positive expression of34cases,negative expression of6cases, the negative expression of SPARCprotein were31cases, SPARCmRNA positive expression of10cases,negative expression of21cases, As the result of Spearmancorrelation analysis, the expression of SPARC protein and SPARC mRNAwas significantly positive, the positive expression of TGFβ1protein were43cases, TGFβ1mRNA positive expression of31cases, negativeexpression of12cases, the negative expression of TGFβ1protein were28cases, TGFβ1mRNA positive expression of11cases,negative expressionof17cases, As the result of Spearman correlation analysis, the expressionof TGFβ1protein and TGFβ1mRNA was significantly positive. Theimmunohistochemistry and in situ hybridization, have high consistency.(1)The expressions of SPARC protein and SPARC mRNA, TGFβ1protein and TGFβ1mRNA and Endoglin protein in lung cancer tissueswere apparently higher than that in adjacent normal tissues, whichpointed out that the high expression of SPARC, TGFβ1and Endoglincould possibly be related to the development of lung cancer;(2)Throughimmunohistochemistry and in situ hybridization, SPARC was associatedwith clinical stage and lymph node metastasis of lung cancer, SPARCmay inhibit lung cancer progress. In early clinical stage, lymph nodemetastasis free, the expression of SPARC increased, MVD was reduced. Both have correlation, the expression of SPARC in lung cancer may berelated to anti angiogenesis.(3)TGFβ1was associated with tumor size,clinical stage and lymph node metastasis of lung cancer. TGFβ1andMVD have significant correlation, TGFβ1was associated with lungcancer progression and angiogengesis.(4)The correlation analysisshowed that SPARC and TGFβ1have negative correlation. As MVDincreased SPARC expression in lung cancer decreased, the expression ofTGFβ1was on the increase, It indicates that on lung cancer angiogenesismay be related to SPARC, TGFβ1, Endoglin interations.
