The medicinal herbs continue to be an important source of bioactive secondarymetabolites. A study was carried out to investigate the potential antioxidant andanti-tumor compounds derived from Euphorbia lunulata Bag. Studied includebioassay-guided fractionation, structural elucidation and preliminary evaluation forantioxidant and anti-tumor activities for pure compounds.Main contents and experimental results are as follows:1. The components in Euphorbia lunulata Bag were extracted by the refluxextraction method and the traditional water extraction method, and extracts weresuccessively fractionalized with petroleum ether, ethyl acetate and n-butyl alcohol toyield crude extracts. The method of SRB and DPPH scavenging rate were used tostudy the pharmacological activity of crude extracts in vitro. The results showed thateffective chemical ingredients consisted in ethyl acetate layer and n-butyl alcohollayer.2. TLC, silica gel column chromatography, gel column chromatography, HPLCand other modern separating methods were used to isolate, purify compounds. AndUV, IR, MS and NMR and other spectroscopic data were used to identify thesecompounds. Twelve compounds were obtained and seven of them were elucidated as(1)methyl4-hydroxybenzoate、(2)4,8-dihydroxy-3,4-dihydronaphthalen-1(2H)-one、(5)1H-indole-3-carboxamidede、(6)1-methylazetidine-2,3-diol、(10)N-[2-(1H-indol-3-yl)ethyl]acetamide、(11)β-sitosterol、(12)N-[3-(4-hydroxyphenyl)propyl]acetamide.3. The antioxidant activity of these compounds was investigated by DPPH scavenging rate, and the anti-tumor activity of them was evaluated by SRB.Compound2and8-12showed a certain degree of apoptosis inducing, cytotoxicactivities or inhibiting the proliferation of tsFT210. Compounds1-2and6-12haveantioxidant activity to a certain extent, compound3-5basically have no antioxidantactivity. Compound8-11have good activity and other compounds have lower activity.In summary, the study discussed the material foundation of anti-tumor activityand antioxidant activity of Euphorbia lunulata Bag, elucidated the main structuretypes of effective components, provided new structure types for drugs havingantioxidant activity and anti-tumor activity, broadened the fields of medicine value ofEuphorbia lunulata Bag, developed medicine value of Euphorbia L andEuphorbiaceous plants, and provided the reliable basis for researching and developingthem.
