Objective:To investigate the clinical significance of Caspase-3and HSP60expression andanalysis the relationship between the expression and the proliferation, apoptosis, anddifferentiation of cells in Astrocytoma by immunohistochemistry.Method useWe consecutively collected tumors from58Patients (mean age,34.6; range,7-65years; male: female,37:21) who had undergone surgery for Astrocytoma at the HuzhouCentral Hospital of the PLA from June2005to December2012. The histopathologicaldiagnosis was made according to the WHO classifieation:4cases were WHO Ⅰ grade:18cases were WHO Ⅱ grade:19cases were WHO Ⅲ grade; and17cases were WHOⅣ grade.Immunohistochemical staining of HSP60and Caspase-3monoclonal antibodywas done using the SP Staining System. SPSS19.0was used to process data with rank sumtest and spearman correlation coefficients test according to date type, considering sensestatistically(P<0.05).Results:1. Immunohistochemistry were done to evaluate HSP60and Caspase-3proteinexpression, which show brown, respectively. The HSP60protein expression had acytoplasmic and nuclear staining pattem, and the Caspase-3had a cytoplasmic staining intumor cells. 2. The HSp60expressed intensity of tumor cells were increase with the histologicalgrade up in Astrocytomas, and the percentage of stained tumor cells from histological grade1to IV was20%,72.2%,89.4%and100%(P <0.01), respectively.3. The Caspase-3expressed intensity of tumor cells were decrease with thehistological grade up in Astrocytomas, and the percentage of stained tumor cells fromhistologieal grade1to IV was90%,77.7%,36.8%and23.5%(P <0.01),respectively.4. Caspase-3expression was reverse correlated with HSP60expression (P <0.01) inAstrocytomas.ConclusionThe Caspase-3and HSP60Protein expression are related to the biological behaviorand differentiation of tumor cell in Astrocytoma. The immunohistochemical examination ofCaspase-3and HSP60expression in Astrocytoma, which have different histological grade,should be help for evaluated the tumor cell malignant degree and have significant todemonstrate the mechanism of the carcinogenesis of Astrocytoma.
