The purple sweet potato extracts are natural extracts extracting from purple sweet potato root, which main activitive constitution is purple sweet potato anthocyanins. It has many kinds of biological activities such as antioxidant, scavenging free radical, reducing blood fat, antibacterial, antimutation. Natural aging climacteric rats were used as model to study the effects of purple sweet potato extracts on climacteric syndrome and related mechanism through observing weight and ovarian tissue morphology of rats, measuring physiological and biochemical indexes as well as gene expression quantity on the molecular level.Firstly,11~12months of age female SD rats which have no regular estrous cycle were used as climacteric syndrome model through observing smear of vaginal cell. They were fed with different dose(50mg/(kgbw·d)、100mg/(kgbw·d)、200mg/(kgbw·d)) of purple sweet potato extracts and soy isoflavone(500mg/(kgbw-d)) to observing the effect of purple sweet potato extracts on climacteric syndrome.The results showed that the purple sweet potato extracts can improves the increasing weight of climacteric rats effectively.Secondly, physiological and biochemical indexes of rats were measured. The results showed the purple sweet potato extracts can increase the index of uterus, vagina and spleen and decrease the level of FSH, LH in serum of climacteric rats significantly, regulating hormone levels, but had no significant effects on E2. The purple sweet potato extracts can improve ovary histomorphology and promote development of all levels of follicles, reduce the number of atresia follicles. The purple sweet potato extracts increased the T-super oxide dismutase (T-SOD), catalase (CAT), decreased methane dicarboxylic aldehyde (MDA) in-serum and liver significantly as well as improving oxidative stress in the body. The purple sweet potato extracts decreased the level of total cholesterol(TC), triglyceride (TG) in serum as well as5-hydroxy tryptamine (5-HT) and activities of monoamine oxidase (MAO) in hypothalamus to improve blood lipid metabolism and vegetative nerve functional disturbance, but had no significant effects on dopamine(DA)in hypothalamus. The estrogen receptor (ER) level in uterus were increased by PSPE significantly.Finally, the real-time PCR experiments on the expression levels of ER-α、ER-β mRNA showed as follows. The expression levels of ER-a mRNA in uterus of climacteric rats were very low and the ER-β mRNA almost had no expression. The purple sweet potato extracts can increase the expression levels of ER-α、ER-β mRNA in uterus of climacteric rats to mediate estrogen signal transduction pathway and promote estrogen play a physiological role in target organs.
