| Objective:To study the quality standards of the Mongolian traditional medicine called rhododendron which provide proof for standard system.Methods:the microscopical identification and TLC were adopted to analyze the leaves and flowers of rhododendron and to determine the contents of the active components including hyperin,quercetin and rhododendron. Then the condition were as follow:the column was Diamonsil C18(150mm×4.6mm,5μm) and the temperature was40℃as well as the velocity was1.0mL/min;then the wavelength were360nm for hyperin and quercetin and299nm for rhododendron.In addition,to determine the contents of water,total ash,acid-insoluble ash,extractives and the heavy metals according to the Chinese pharmacopoeia(2010editon).Results:Pollen grain and sclereid were found in the flowers oscopical identification and glandular hairs, stoma,calcium oxalate crystals,nonglandular hairs,wood fiber,duct,bast fibre as well as glandular scale and so on in the leaves were researched.Then the results of TLC showed that the spot of the samples were in accordance with the ones of the controlled substances.Furthermore,it was found that the results of HPLC were as follow:the regression equation of hyperin was Y=2062266.9285X-11243.3118, r=0.9997,whose linear range was0.1608～0.9649μg and the average percentage of recovery was99.94%; the regression equation of quercetin was Y=3019428.5714X+9805.8667, r=0.9996,whose linear range was0.0181～0.1086μg and the mean recovery rate was99.17%;the regression equation of rhododendron was Y=3805506.4875X-800.0311, r=0.9996,whose linear range was0.00470～0.02805μg and the average recovery was99.36%.Otherwise,the other examinations accords with the regulation of Chinese pharmacopoeia published in2010.Conclusions:The methods of microscopical identification,TLC and HPLC could be used to control the quality quickly and easily that offered the scientific proofs for establishing the quality standard of Rhododendron smsii planch. |
