Objective:The study observes the effects of electro-acupuncture at Baihui, Dazhui, Shenshu (acupoints) combined with aerobic exercise on the expression of ICAM-1and related factors of cognitive function in rats model of chronic ischemic cognitive impairment. The possible mechanism is investigated on electro-acupuncture combined with aerobic exercise to improve the cognitive impairment of chronic ischemic. Comparing the differences of treatment effects among the electro-acupuncture combined with aerobic exercise and single aerobic exercise or single electro-acupuncture, the study provides more experimental evidence of clinical application on treatment of chronic ischemic cognitive Impairment by using electro-acupuncture combined with aerobic exercise.Methods:In this experiment, the60healthy male SD rats were randomly divided into6groups,10rats in each group. Such as the blank group (group A), sham operation group (group B), ischemia group (C group), aerobic exercise group (group D), electro-acupuncture group (group E), combined treatment group (group F). In addition to group A and group B, the other groups were made the model of chronic cerebral ischemia by permanent bilateral common carotid artery ligation. And the group B separated the bilateral common carotid artery without ligation or cut. After8weeks treatment of electro-acupuncture at Baihui, Dazhui, Shenshu combined with aerobic exercise, the cognitive function was tested by Morris water maze in rats, the morphology of the Nissl bodies was observed by Nissl staining, the positive expression of ICAM-1in hippocampus of rats was detected by immunohistochemistry, and the content of TNF-a, IL-6were detected by ELISA. Results:(1) After modeling, the water maze tests in rats escape latency were prolonged. Compared with the blank group, the difference was statistically significant (P<0.01), and this indicated the model was successful. After8weeks intervention, the escape latency was significantly shortened in the group D, E, F. The difference was statistically significant compared with the blank group (P<0.05). The water maze performance evaluation in group F was better than group D or group E.(2) The rats in group A and group B showed a large number of intact structure and regular arrangement of Nissl bodies, and the nucleolus is clear without edema or inflammatory cell infiltration. The rats in group C showed a small amount of Nissl bodies, and the cell nuclear volume nosedived. These were a large number of hippocampal pyramidal cells apoptosis in this group. The rats in group D, group E, group F showed a small amount of cell apoptosis and Nissl bodies were rich. And the apoptosis of pyramidal cells in hippocampus of rats with significant difference compared with ischemia group.(3) It showed that electro-acupuncture and aerobic exercise could reduce the expression of ICAM-1and related factors in rats of chronic cerebral ischemia in group D, group E, and group F. The difference was statistically significant compared with the blank group (P<0.05). The expression of ICAM-1and related factors in group F was lower than group D or group E, and the difference was statistically significant (P<0.05). There was no significant difference between group D or group E (P>0.05).Conclusion:(1) Electro-acupuncture at Baihui, Dazhui, Shenshu (acupoints) and aerobic exercise can improve the cognitive function, learning and memory ability of chronic ischemic type cognitive dysfunction in rats. These methods can improve the pathological injury degree of hippocampus as well. Comparing the difference of treatment effects, the electro-acupuncture combined with aerobic exercise is better than single aerobic exercise or single electro-acupuncture.(2) It showed that electro-acupuncture and aerobic exercise could reduce the expression of ICAM-1and related factors, and reduce the damage of the cognitive function of chronic cerebral ischemia, and improve learning and memory ability of rats. The electro-acupuncture combined with aerobic exercise is better than single aerobic exercise or single electro-acupuncture. This illustrates the cognitive dysfunction could be remedied by electro-acupuncture combined with aerobic exercise to control the inflammatory cytokines, relieve the inflammatory injury of nerve cells, and to relieve the rat model of cerebral damage.
