Objective:This study taking FD rats as research objects, to observe the influence of electroacupuncture at ST36on the gastric emptying,small intestine propelling rate, the ultrastructure of ICC and expression of protein Cx43in FD rats, and to explore clarify whether electroacupuncture at ST36regulating gastrointestinal dynamics of FD rats with the change of the ultrastructure of ICC and expression of protein Cx43, so as to clarify the mechanism of electroacupuncture at ST36on the gastrointestinal dynamic in FD rats.Method:48SD rats were randomly divided into4groups:A.normal group, B.model group, C. Electroacupuncture group, D. Duo-pan-li-tong group. Making model by clamping tail method, after making model, the electroacupuncture group rats receive electroacupuncture at ST36treatment for10days, while the other3groups of rats receive binding. Duo-pan-li-tong group receive turbid liquid for10days according to the dose of1ml/100g·d, while the other3groups of rats receive normal saline for10days according to the dose of1ml/100g·d. There is no food but water for four groups of rats after treatment, and receive semisolid paste according to the dose of1ml/100g. After recording30minutes, experimenting the gastric emptying and small intestinal propulsion, and observing the ultrastructure of ICC by transmission electron microscope, and observing the expression of protein Cx43by immunohistochemical method.Results:1.Comparison of weight and appetite:compared with the normal group, the weitht and appetite of the other3groups were decreased, the difference was statistically significant (P<0.05or P<0.01). After treatment, compared with the model group, the weitht and appetite of the other3groups were increased, the difference was statistically significant (P<0.01). When compared with the electroacupuncture group, the weight and appetite of the Duo-pan-li-tong group were higher than the electroacupuncture group, the difference was statistically significant (P<0.01). 2.Comparison of gastric residual rate:compared with the normal group, the gastric residual rate of the model group were increased, the difference was statistically significant (P<0.05orP<0.01). After treatment, compared with the model group, the gastric residual rate of the electroacupuncture group and the Duo-pan-li-tong group were decreased, the difference was statistically significant (P<0.01). When compared with the electroacupuncture group, the gastric residual rate of the Duo-pan-li-tong group has no significant difference(P>0.05).3.Comparison of small intestinal propulsion:compared with the normal group, the small intestinal propulsion of the model group were decreased, the difference was statistically significant (P<0.05). After treatment, compared with the model group, the gastric residual rate of the electroacupuncture group and the Duo-pan-li-tong group were increased, the difference was statistically significant (P<0.05or P<0.01). When compared with the electroacupuncture group, the small intestinal propulsion of the Duo-pan-li-tong group has no significant difference(P>0.05).4.Comparison of the ultrastructure of ICC:the number and ultrastructure of ICC were significantly changed in model group, the connections between the surrounding cells and smooth muscle were decreased, and most the mitochondria has been swollen. Compared with the model group, the number and ultrastructure of ICC and the connections between the surrounding cells and smooth muscle in electroacupuncture group have been significantly improved.5.Comparison of integral optical density of Cx43:compared with the normal group, the integral optical density of Cx43in model group were decreased. Compared with the model group, the integral optical density of Cx43in the electroacupuncture group and the Duo-pan-li-tong group were increased, the difference was statistically significant (P<0.01).Conclusion:1.After making models, the appetite of rats were decreased, and the weight of rats were decreased or increased slowly, and there is no ulceration or other organic diseases in stomach tissue by naked eye, which illustrates that the making model was successfully.2.Compared with the normal group, the difference of weight, appetite, gastric residual rate, small intestinal propulsion in the electroacupuncture group and the Duo-pan-li-tong group was statistically significant, which illustrates that both the electroacupuncture and Duo-pan-li-tong can treat the FD rats by regulating gastrointestinal dynamics.3.Both the electroacupuncture and the Duo-pan-li-tong can increase the number and ultrastructure of ICC and the expression of protein Cx43, so that they can promote the recovery of gastrointestinal dynamics.4.Electroacupuncture at ST36promote the recovery of gastrointestinal dynamics, this may be related with the change of the ultrastructure and the function of ICC and promoting the expression of protein Cx43.
