Ants have cured of disease in our country since ancient times. Chinese pharmacopoeia records ants mainly for liver and kidney, tonifying kidney and attending kidney deficiency,dizziness and tinnitus, spermatorrhea, impotence, rheumatism, hemiplegia stroke and other diseases. Ants for medicinal research,Chinese scholars mostly follow the characteristics of Chinese medicine research, i.e.the experience of medicine, for clinical first, and then basic experiment. The most famous is the pioneer of Contemporary ants therapy Professor Zhicheng Wu, he combines traditional Chinese medicine therapy and clinical academic thoughts, proposed ants as sovereign medicine to treatment rheumatoid arthritis, ankylosing spondylitis, hepatitis B and hepatitis B virus. The efficacy of the medicine is the synergistic effect of chemical constituents, a link must exist between the two. At present, the research on the correlation of chemical constituents and pharmacological activity is scarce, leading to the application of the ants be limited.In order to systematically and thoroughly studied ants pharmacodynamic activity based, this experiment from attending acute or chronic hepatitis compound GanLaoXin sovereign medicine red ants (Tetramorium.sp) as the starting point, first of all we mainly used the means of silica gel, ODS, Sephadex LH-20, etc. column chromatography to isolate the chemical constituents of the90%ethanol extract of Tetramorium.sp and gained9compounds. The structure of eight compounds was determined by nuclear magnetic resonance (NMR) and mass spectrometry (MS) and other spectroscopic techniques, wherein the compound3,6and7separated from the ants for the first time.Second, the volatile oil of10different origin ants was extracted by steam distillation. GC-MS analysed the differences of the volatile oil contained in the ants, which combined with subsequent macrophage inflammatory factor experiment to explore the relationship between the chemical constituents of volatile oil and the anti-inflammatory activity.Also for the red ants were studied by chemical, physical means to confime the presence of triterpenoids, and its quantitative determined the material content of triterpenoids in ants is about2-3%.LPS (Lipopolysaccharides) can induce RAW264.7murine macrophage inflammatory response and release of tumor necrosis factor TNF-α and interleukin-inflammatory cytokines such as IL-1β. This subject determined the content of inflammatory cytokines by ELISA (enzyme linked immunosorbent assay),and preliminary explored the inhibition of these two inflammatory cytokines by methanol/acetone crude extracts, ants volatile oil and four monomers compounds. The results showed that the methanol/acetone crude extracts, essential oils, monomer compounds have a certain degree of anti-inflammatory activity. Combined with the LC-MS spectra of10batches of crude extracts and volatile oil GC-MS data, PLSR explored the spectrum peaks associated with the anti-inflammatory activity, as well as its contribution to the anti-inflammatory activity.
