| Ligusticum jeholense Nakai et Kitag. is the plant of Ligusticum in Umbelliferae, Its root and rhizome are a Chinese herbal medicine used as a drug for treatment of fever, cold, headache and rheumatism. And it is specified in every Pharmacopoeia of the P.R. China.The chemical constituents from EtOH extract of the root and rhizome of L.jeholense. were systematically researched in this thesis and 8 compounds were isolated. Their chemical structures were identified by spectral methods, including levistolide A(LJG-1), xiongterpene(LJG-2), linoleic acid(LJG-3), linum cerebroside(LJG-4), ferulic acid(LJG-5), sucrose(LJG-6), daucosterol(LJG-7),β-sitosterol(LJG-8).Among these 8 compounds 3 and 4 compounds were isolated for the first time from L.jeholens. 1~2 and 6~7 compounds were isolated for the first time from Ligusticum.The chemical constituents from EtOH extract of the aerial part of L.jeholense. were researched and 7 compounds were isolated. Their chemical structures were identified by spectral methods, including kaempferol-3-O-β-D-galactoside(LJD-1), quercetin-3-O-β-D-galactosid(eLJD-2), kaempferol -3-O-(2",4"-di-E-p-coumaroyl) -α-L-rhamnosid(LJD-3), psoralen(LJD-4), sucrose(LJD-5), daucosterol(LJD-6),β-sitosterol(LJD-7).Among these 7 compounds 1~4 compounds were isolated for the first time from Ligusticum. 5~7 compounds were isolated for the first time from the aerial part of L.jeholense.The anti-oxidative activity of the underground part and the aerial part of L.jeholense. was evaluated by DPPH free radical scavenging method. Althouth the results indicate that the underground part and the aerial part both display scavenging activity, the aerial part has 3.29 times stronger scavenging activity than the underground part.For the further studying anti-oxidative activity of the aerial part, the anti-oxidative activity of different polar solvent extracts of EtOH extract of the aerial part were also evaluated by DPPH free radical scavenging method. The results show that the different polar solvent extract all display scavenging activity, but the activities are different. The order is that EtOAc extract>n-BuOH extract>CHCl3 extract>P.E extract. The EtOAc extract have stronger scavenging activity than Vitamine E among these extracts.The EtOAc extract of the aerial part was analyzed by LC-MS(n) and we preliminary identified the structures of 7 chemical constituents. Among these 7 compounds ,4 compounds haven't been published and 3 flavones have been known . This study made for further realizing and isolating its anti-oxidative active constituents.The volatile oil of the root and rhizome of L.jeholense. was analyzed by GC-MS. And we identified the structures of 17 chemical constituents, accounting for 23.56% of the total peak area. This research is beneficial to understand the chemical constituents of the volatile oil of the root and rhizome of L.jeholense and plants of Ligusticum. |
