BackgroundAs a type of coronary heart disease,acute myocardial infarction (AMI)is more serious, and its acute onset and high fatality rate threatened people’shealth seriously. At present the diagnosis of AMI is determined by theclinical manifestation, characteristics of electrocardiogram and the dynamicchanges in serum biomarkers. it has been an important subject of ourresearch to earlier diagnosis and prevention of acute myocardial infarction。With the completion of human genome project and the development ofmicroarray technology, virulence genes of many single gene diseases werefound and applied to the diagnosis and treatment of diseases. As a kind ofdisease affected by a variety of genetic and environmental factors, thepathogenesis of AMI is still not entirely clear.Participate in the AMIprocess. The study investigates which specific genes participate in the AMIprocess by the differential expression analysis of AMI peripheral bloodRNA, laying the foundation for the early gene diagnosis of AMI in future.ObjectiveThe microarray analysis is performed for peripheral blood RNA ofAMI patients and healthy people to screen the differential expression geneswhich participate in the AMI development process and find the molecular markers for AMI early diagnosis.MethodMicroarray analysis of differentially expressed in peripheral bloodRNA in patients with acutemyocardial infarction.The research objects: Wechose12cases of AMI patients and12cases of CHD patients in ourhospital for treatment in2012.06-2014.08as comparative groups. Weextracted mRNA in peripheral blood leukocytes, then reversed andtranscribed them into cDNA. We chose3cases of AMI patients and3casesof comparative group patients to detect their gene chip. Other samples wereused for validating the gene chip results of RT-PCR detection. After that,we selected CYP4F3, IL13RA1, USP25randomly in chip results to do theRT-PCR detection, which were on behalf of the high gene expression,indifference and low gene expression group respectively. Line of real-timefluorescent quantitative PCR analysis these gene expression differences indifferent groups.ResultAccording to the results of gene chip detection,559genes showedabnormal expression in AMI patient group, of which288high geneexpression and271low expression (the results according to the p-value0.05, or less|logFC|>1). GO analysis showed that these genes belong tofrom128cellular_component. They participated in521biological_processand acted151molecular_function; KEGG analysis involved in107genetic pathways. Fluorescence quantitative PCR results: CYP4F3gene expressionin patients with AMI is higher than that of the comparative group (1.62±0.27, p=0.27), while USP25gene expression in patients with AMI is lower(0.56±0.07, p=0.07), and gene IL13RA1expressed no difference in AMIpatient group and the comparative group (1.11±0.35, p=0.35).ConclusionThe result of gene chip analysis is credible. There are differences of RNAin the peripheral blood between patients with AMI and healthy people.CYP4F3and USP25may be genetic markers for diagnosis of acutemyocardial infarction.
