Subject:To find reasons of essential hypertension (EH), clarify the NPPA gene polymorphism in EH patients of Korean Chinese and Original Chinese nationality in Yanbian.Methods:1. Total 140 patients of EH and 156 healthy applicants were included. EH patients were divided into Korean Chinese group (n=67) and Han nationality group (n=73), healthy applicants were also divided into Korean Chinese group (n=67) and Han nationality group (n=89).2. The samples were collected to extract genomic DNA.3. PCR was performed to analyze target mRNA.4. Genotype were identified by rs2270915 rs5065 gene polymorphism with specific restriction enzyme. Gene sequencing was done using the PCR product according the Sanger’s method.Results:1. In Korean Chinese, the rs2270915 locus showed 67%,24%,9% gene frequency of AA, AG, and GG genotype in EH samples. And it showed 64%, 34%,1% in healthy applicants. There was no statistical significant between two groups (P=0.087). In Han nationality, the rs2270915 locus showed 62%,21%, 18% gene frequency in AA, AG, GG genotype in EH patients. And it showed 54%, 43%,3% in healthy applicants. It has significant differences between two groups (P=0.001). In Korean Chinese, the frequency of A and G allele were 77% and 23% in EH group, and they were 81% and 19% in healthy applicants. It has no significant differences between two groups (P=0.436). In Han nationality, the frequency of A and G were 72% and 28% in EH group, and they were 75% and 25% in healthy applicants. There was no significant differences between two groups (P=0.494). According to the dominant module, the genotype frequency about AA+AG and GG were not significantly related with SBP (P<0.05),but were significantly related with DBP (P<0.05) in Korean Chinese, while in the Han nationality, there was significantly difference between AA+AG and GG genotype with gene frequency distribution, SBP and DBP (P<0.05).2. In Korean Chinese, rs5065 locus showed 52%,45%,3% gene relation frequency with AA, AG, GG genotype in EH group; and showed 69%,27%,4% gene relation frequency in healthy applicants group. There was no statistical significant between two groups (P=0.094). In Hari nationality, rs5065 locus showed 62%,30%,8% gene relation frequency with AA, AG, GG genotype in EH group; and showed 62%,37%,1% gene relation frequency in healthy applicants group. There was no statistical significant between two groups (P=0.061). In Korean Chinese, the frequency of A and G allele were 75% and 25% in EH group, and 82% and 18% in healthy applicants group. There was no statistical significant between two groups (P=0.138). In Han nationality, the frequency of A and G allele were 77% and 23% in EH group, and 80% and 20% in healthy applicant group. There was no statistical significant between two groups (P-0.428).3. Total cholesterol (TC), alcohol, BMI were risk factors of EH in Korean Chinese, while the rs2270915 AA+AG (OR:3.612,95% CI:1.467-8.891, P=0.05), alcohol, TC, and low density lipoprotein cholesterol (LDL) were risk factors of EH in Han nationality.Conclusions:1. The genetic polymorphism of NPPA rs2270915 was related with risk of EH in Han nationality, but not in Korean Chinese in Yanbian area.2. Neither Han nationality nor Korean Chinese showed significant relationship between genetic polymorphism of NPPA rs5065 and the risk of EH.3. NPPA rs2270915-AA+AG genotype is one of the risk factor of EH in Han nationality.
