| Object ive:To study the impact of sesamin rabbit aortic endothelial cells, the expression of STAT3 explore sesamin anti-atherosclerotic mechanisms.Methods:The rabbit aortic endothelial cells subculture, which is used in experiments in logarithmic growth phase cells, and divided into four groups according to the experimental needs, at room temperature 37℃,5% CO2 conditions under each experimental group placement cultured in an incubator: ①normal control group:DMEM culture medium; ② ox-LDL group:ox-LDL100mg/L culture 24h; ③AG490+ox-LDL groups:cells were treated with 50μmol/L of AG490 for 24h after joining ox-LDL (final concentration of 100mg/L) cultured 24h;④sesamin+ox-LDL group:cells were treated with 60μmol/L sesamin culture 24h after adding ox-LDL (final concentration of 100mg/L) for 24h again. Each experimental group treated as described above, continues in serum-free culture medium DEME,37℃,5% CO2, humidified incubator environment cultured 24h, Detection of rabbit aortic endothelial cells mRNA levels of STAT3 by RT-PCR method. Results:RNA expression of STAT3 ox-LDL group was significantly higher than the control group, there were significant differences P<0.05, sesamin+ox-LDL group, AG490+ox-LDL group STAT3 RNA expression was significantly higher than the control group, there were significant differences P<0.05, sesamin+ox-LDL and AG490+ox-LDL which there was a significant difference P<0.05.Conclusion:sesamin, mRNA expression of AG490 inhibits STAT3. Sesamin inhibition greater than AG490, with anti-atherosclerosis effect. |
