| Objective:Wnt/β-catenin is one fundamental signaling system that direct cell proliferation, cell polarity, and cell fate determination.Recent studies show that wnt/β-catenin signal pathway has a key role in hepatic fibrosis development,which suggests that it may be the new drug effects in liver fibrosis.Ganfukang is compound traditional Chinese medicine which is developed from our laboratory after years of experiments.It has to improve liver function, reduce inflammation and steatosis, inhibit fibrosis and other effects.Dan shen as anti-fibrosis of single Chinese herb is mainly to improve the micro-circulation, protect the liver cells, reduce the inflammatory response, inhibit the synthesis of collagen, and promote the liver fibrosis reabsorbed.A rat model of fibrosis is induced by CCL4 for 8 weeks.Dan shen and ganfukang is respectively injected into the rat model till the fourth week to the eighth week,and set up the control group:dan shen group, GFK control group.We eviscerate the rat model liver and observe the morphological structure by HE staining, detect the expression of the targets genes.We observe whether ganfukang and dan shen is pro-fibrogenic liver toxicity or not,discussing how two drugs effect wnt/β-catenin pathway,comparing the respective effect of dan shen and ganfukang on wnt/β-cateninin hepatic fibrosis.Methods:Healthy SD rats formed 6 groups randomly:dan shen treatment group;GFK control group;dan shen control group;normal control group;model group;GFK treatment group,every group is ten rats.Except the control group,other rats were given CCL4 subcutaneously(5ml/kg) until the eighth week and then sacrificed.The normal groupã€the dan shen control group and the GFK control group was given saline subcutaneously for 8 weeks.The drug groups and the two treatment groups used drug combination from the fourth week until the eighth week.All the drugs were dissolved in the saline(10ml/kg).The rats were sacrificed at the end of the eighth week for making paraffin section and molecular biological examinations.Results:Observe the mental state of the rats,feeding and movement in each group.When the rats were sacrificed,we observe the liver of the rats every group.The pathological change of the model group was serious.The GFK treatment group had been improved obviously. Immunohistochemistry showed that GFK down-regulated expression of alpha-smooth muscle acin(α-SMA).The expression of Wnt1,Wnt3 a were significantly up-regulated in the model group,while down-regulated in the GFK treatment group compared with the model group.They were at low lever and no statistical significance in the three control groups.But the dan shen treatment group was also no statistical significance with the model group.In the Western blot result,β-catenin protein in the model group increased and decreased in GFK treatment group,While the expression of the control groups were at a relatively low level.1.Dan shen and ganfukang have no effect on the wnt classic pathway.2.Ganfukang can inhibit the activation of the wnt/β-catenin to improve liver function and alleviate fibrosis.3.The ganfukang’s fibrosis degree of inhibition rats liver is more ideal than the dan shen.Conclusions:... |
