Objective:Observing micro-skin graft for external use of chitosan/glucomannan (CS/KGM) composite membrane is whether can be in favour of angiogenesis and epithelium, and provide skin with continuous and good growth environment.It will help look for a kind of wound covering which is safe, convenient of storage and transportation, easy to spread, and solve the problems of the short protection time and poor quality of cell-free pigskin for clinic.Methods:Animal experiment used its own control method.Three days before surgery, we used preoperative hair removal agent in experimental rats around the small of the back on both sides of the symmetrical parts.The hair removal area was about 6cm*6cm, and was protected with the external use glycerin.According to the random number table, The will be on both sides were randomly divided into experimental group (group A) and control group (group B).Firstly, we gave 3% pentobarbital sodium injection(lmg/kg) in the experimental rabbit ear margin for intravenous anesthesia.Then we used methylene blue symmetric to draw 4cm*4cm operation area in the small of the back left and right sides of the hairremoval parts.After conventional iodine volts disinfection and spread towels, we used a roller blade to get blade thick skin in the symmetry operation area.Then, we made micro-skin by standby transplant procedures. We made a model of about 4cm*4cm area of full-thickness skin defects.Finally, the micro-skin in accordance with 1:10 scaled was replantated into the original wound.Group A and group B were respectively covered with chitosan/glucanmannan composite membrane and cell-free pigskin, interrupted suture and tied over dressing. We set the operation after lw,2w,3w,4w and 5w as the experimentation of wound healing based on point in time.We would cut each about 1cm*1cm thick skin all in the experimental rabbit around the small of the back on both sides of the symmetrical wound at each time point to do HE staining and immunohistochemical detection of PCNA and CD31, observed the chitosan/konjac glucomannan composite membrane and acellular pigskin appearance, adhesion, wound healing and wound healing, so as to evaluate the wound repair quality comprehensively.ResultsrWe found that the wound healing rate of the experimental group and the control group was not significant in the first week(p>0.05).The experimental wound healing rate was higher than the control group in the second to fourth week, and the differences between the two groups were significant (p<0.01); The wound of the experimental group and the control group was completely healed in the fifth week.There were no significant differences between the two groups (P>0.05).Immunohistochemical staining was used to detect the expression of CD31 and PCNA.The expression of CD31(microvessel density) in the experimental group were significantly higher than the control group in the first to fifth week(P<0.01).The positive expression rate of PCNA experimental group was higher than the control group in the first and second week (P<0.01); The positive expression rate of PCNA in the experimental group was lower than the control group in the third and fourth week (P<0.01); Because the experimental group and the control group were healed without wound in the fifth week, we did not compare the positive expression rate of PCNA again.Conclusions:Chitosan/konjac glucomannan composite membrane compared with cell-free pigskin, It is more conducive to promote wound endothelial cells and epithelial cells differentiation, proliferation, migration, angiogenesis and accelerate epithelization, so as to improve the quality of wound healing.Chitosan/glucomannan composite membrane has good compatibility with wound tissue and antibacterial activity.It makes micro-skin and the wound firmly attached, and it is a good neighbourhood to micro-skin survival and wound healing.The chitosan has antibacterial activity, hemostatic effect, and water absorption.The glucomannan can increase the mechanical strength of the wet state of the chitosan and it maakes the composite membrane attached to the wound and peripheral tissue suture easily.As the coverage of micro-skin graft, we believe that chitosan/glucomannan composite membrane is better than cell-free pigskin...
