Brain-derived neurotrophic factor(BDNF) is attributable to nerve growth factor family, Its wide range of biological effects, Can maintain brain, spinal ganglia, cerebral cortex, hippocampus, striatum survival of developing neurons and support neurons in vitro survival and neurite growth. In this study, immunohistochemical techniques were applied to investigate the expression of rabbit syringomyelia surrounding tissue Tropomyosin receptor kinase B(TrkB) and P75 neurotrophin receptor after BDNF intervention. Simultaneous sensory and motor function score. Proposed new method to observe the Therapy for scoliosis associated with cervical syringomyelia in rabbit by BDNF, And discuss the possible mechanism of BDNF promote repair of spinal cord lesions.Objective:Analysis the molecular biological changes in tissue surrounding the syringomyelia and the changes in sensory and motor functions because BDNF’s participation in treatment. For the purpose of doing basic research on the application of BDNF in the future.Methods:1.Take 50 healthy Japanese big ear rabbits under direct vision Kaolin within 60 ul spinal injections into the seventh cervical spine segment, to production non -communicating syringomyelia model.2. At time points of 4 weeks、8weeks、12weeks after surgery formodified Tarlov score and the score of pain and temperature sensation.3. At time points of 4 weeks、8weeks、12weeks after surgery for X-ray, MRI scan.4. The successful establishment of a non-communicating syringomyelia associated with scoliosis model is divided into blank group,control group and the treatment group.5.The control group under Stereo positioning instrument saline within 60 ul spinal injections into the seventh cervical spine segment internal tissues. And the treatment group under under Stereo positioning instrument BDNF(10ng/ml) within 60 ul spinal injections into the seventh cervical spine segment internal tissues.6. At time points of 4weeks、8weeks、12weeks after the treatment for pain and temperature ratings and improvement of Tarlov score.7. At time points of 8weeks、12weeks after treatment immunohistochemical technique were used to detect BDNF TRB, P75 receptor expression changes. Immunohistochemical results using computer aided Image analyzer (Metamorph Image Sys-tem (4.6)) for quantitative analysis.Results:1.To 12 weeks after, There’re a total of 30 in 50 continuous observation of experimental animals can only be observed syringomyelia. MRI showed cervical syringomyelia main living under, syringomyelia all show the limitations of the central tube expansion, sizes.30 of 29 rabbits had occurred different extent of cervical thoracic scoliosis.2.Treatment groups at different time points around the Syringomyelia tissue TRB and P75 was stronger than in the control group and the control group.3.Treatment group at different times Tarlov scores improved obviously superior to control group and blank group.4.Treatment group at different times Pain and temperature ratings improved obviously superior to control group and blank group.Conclusion:1.BDNF at different time points after intervention treatment can improve the damaged tissue around the TRB, P75 expression.2.BDNF at different time points after intervention treatment can obviously improve experimental animal movement, sensory function.
