Objective:Research of soyasaponins on D-galactose induced anti-aging effect of mice and its mechanismMethods:50 Kunming mice were divided into the blank control group, experimental D-galactose model group, soyasaponins low dose(L) group(50mg/kg), high dose(H) group(100mg/kg) and vitamin E group,10 rats in each group. The blank control group were treated by subcutaneous injection of 0.7% NaCl solution every day,300mg/kg each time, and the remaining group of mice were injected in the same place using the same volume of D-gal, injected every day, for 7 weeks. With the injection of D-gal, L group, H group and vitamin E group were gavaged with different dose respectively, and the dose were 50mg/kg, 100mg/kg,50mg/kg. The blank control group and model group were gavaged with an equal volume of 0.7% NaCl solution. The last gavage were fasting but water, taking the blood, liver and heart 12 hours later.8-hydroxy-2 -deoxyguanosine (8-OHdG), advanced glycation end products (AGEs) detection of senility indexes were undertaken after serum was obtained ; Also the biochemical indicator of superoxide dismutase (SOD), malondialdehyde (MDA), monoamine oxidase (MAO); the immunological index of tumor necrosis factor alpha (TNF-a), interleukin-1 (IL-1β), nitric oxide (NO), prostaglandin (PEG-2) were detected. Liver and heart sections were stained with HE and embedded in paraffin for morphological detection. Small patch of liver and heart were taken and detected by means of transmission electron microscope imaging detection. Part of liver and heart homogenate were taken and detected the biochemical indexes of SOD, MDA, MAO, NO, iNOS; then made detection and NF-κB pathway of COX-2, HO-1, iNOS therapeutic targets by Western Blot method.Results:1、Detection of the serum of mice:compared with the blank control group,8 - hydroxy deoxyguanosine and advanced glycation end products (AGEs) activity of D-gal model group were increased (P<0.01), compared with the D-gal model group,8 - hydroxy deoxyguanosine and advanced glycation end products (AGEs) activity of the soyasaponins treated group were significantly decreased (P<0.01); compared with the blank control group, SOD activity of D-gal model group were decreased (P<0.01), compared with the D-gal model group, SOD activity of the soyasaponins treated group were significantly increased (P<0.01); compared with the blank control group, the MDA, MAO activity of D-gal model group were significantly increased (P<0.01); compared with the blank control group, the contents of TNF-a, IL-1(3, NO and PEG2 of D-gal model group were significantly increased (P<0.01), compared with the D-gal model group, the contents of TNF-a, IL-1β, NO and PEG2 of soyasaponins treated group were decreased (P<0.01).2、Detection of liver and heart tissue of mice:compared with blank control group, SOD activity of D-gal model group was decreased (P< 0.01), compared with D-gal experimental model group, SOD activity of soyasaponins treated group was increased significantly (P< 0.01);compared with blank control group, MDA, MAO activity of D-gal model group were increased significantly (P< 0.01), compared with D-gal experimental model group, MDA, MAO activity of soyasaponins treated group were significantly decreased (P< 0.01);compared with blank control group, the contents of iNOS, NO in D-gal model group was obviously increased (P< 0.01), compared with D-gal experimental model group, the contents of iNOS, NO of soyasaponins treated group were obviously decreased (P< 0.01); Liver and heart tissue HE staining showed that soyasaponins played a protective role on aging mice caused by D-gal; liver and heart tissue morphology showed that soyasaponins played a protective role on aging mice caused by D-gal. Important therapeutic targets of liver and heart injury were tested by Western blot, compared with blank control group, COX-2, HO-1 and iNOS expression of D-gal model group were increased significantly, compared with D-gal group, COX-2, HO-1 and iNOS expression of soyasaponins treated group were significantly reduced; detection of NF-κB and p-NF-KB protein in upstream signal pathways, compared with blank control group, NF-κB and p-NF-KB expression of D-gal model group were increased significantly, compared with D-gal experimental model group, NF-κB and p-NF-KB expression of soyasaponins treated group were decreased obviously.Conclusion:Soyasaponins has the effect of anti-aging on mice induced by D-galactose. The anti-aging mechanism may be related to the improvement of body’s antioxidant capacity and the reduction the body’s inflammatory response. The anti-inflammatory effect of soyasaponins is related to the regulation of NF-κB.
