Objective:To study the effect of soyasaponin on anti-oxidation of kidney in the mice induced by D-galactoseMethods:Sixty male SPF Kunming mouse were randomly divided into six groups. Except for the control group, groups of D-galactose (D-gal) model, soyasaponin low dose(SS25), soyasaponin middle dose (SS5O), soyasaponin high dose(SS100) and vitamin E (VE) were respectively administered 300mg/kg D-gal, 25mg/kg SS plus D-gal,50mg/kg SS plus D-gal, 100mg/kg SS plus D-gal,50mg/kg VE plus D-gal once daily for a period of 7 weeks. After the final gastrogavage, blood was centrifuged (2000rpm,15min) 2 h later and serum samples were stored at-40℃. Then dissected the kidney by killing mice. The method of this research as follows:1. 8-hydroxydeoxyguanosine(8-OHdG) in blood serum was determined by ELISA.2. Kidney sections were embedded in paraffin and stained with HE for pathologic morphological observation;3. The oxidation indicator of superoxide dismutase(SOD), malondialdehyde(MDA), glutathione peroxidase(GSH-PX), hydrogen peroxiase, nitric oxide (NO), inducible nitric oxide synthase(iNOS) of kidney homogenate were measured by colorimetric methods;4. Activities of Na+K+-ATPase, Ca2+Mg2+-ATPase and oxidation indicators of Mn-superoxide dismutase (MnSOD), malondialdehyde (MDA) of mitochondrion was assayed by colorimetric methods; 5.The expression patterns of the rapeutic targets COX-2, HO-1 were assayed by Western Blot.Results:1.Detection of the serum of mice by ELISA:compared with the blank control group, the content of 8-OHdG of D-gal model group were significantly increased (P<0.05), compared with the D-gal model group, the concentrations of 8-OHdG of all SS groups were significantly decreased (P<0.05).2. The pathologic morphology results of renal tissue shows that:Size and shape of glomerulus was normal, abnormalities of renal tubule was not found in the control group. Compared with the control group, D-gal model group was found that the glomerulus obviously atrophy, part of the renal tubular epithelial cell swelling, and lumen narrowing; Compared with D-gal model group, renal tubular epithelial cell of SS25 group was obviously swelling, lumen narrowed considerably, and glomerulus was not seen obvious atrophy; SS50 group was found hyaline degenaration; Renal tubular epithelial cells of SS100 group were not seen obvious swelling, abnormalities of the size and shape of glomerulus and lumen were also not found.3. The oxidation indicator in kidney tissue of mice:compared with the control group, activities of SOD, GSH-PX, CAT of D-gal model group was significantly decreased (P<0.05), compared with the D-gal model group, activities of SOD of all SS groups and activities of GSH-PX, CAT in SSI 00 group was enhanced significantly (P<0.05);compared with the control group, content of MDA in D-gal model group were increased significantly (P<0.05), compared with the D-gal model group, contents of MDA in all SS were significantly decreased (P<0.05);compared with the control group, the level of NO and iNOS in D-gal model group was obviously increased (P<0.05), compared with the D-gal model group, the contents of NO and the activities of iNOS in all SS groups were declined significantly(P<0.05);4. Detection of kidney mitochondrion of mice :ompared with the control group, activity of Na+K+-ATPase, Ca2+Mg2+-ATPase, MnSOD in D-gal model group was significantly decreased (P<0.05), compared with D-gal model group, activities of Na+K+-ATPase, Ca2+Mg2+-ATPase of all SS groups and MnSOD of SS100 group was increased significantly (P<0.05);compared with control group, content of MDA D-gal model group were increased significantly(P<0.05), compared with D-gal model group, contents of MDA in SS100 group were decreased significantly(P<0.05).5. Important therapeutic targets of kidney injury were determined by Western blot:compared with the control group, expression of HO-1 and COX-2 of D-gal model group were promoted significantly(P<0.05), compared with D-gal experimental model group, expression of HO-land COX-2 of all SS groups were decreased obviously(P<0.05).Conclusion:Soysaponin has delaying effect on kidney aging of the mice induced by D-gal;Soysaponin may improve body’s antioxidant capacity by reducing free radicals damage to the body;Soyasaponin has anti-aging effect on mice induced by D-galactose,whose mechanism may be related to the elevated antioxidant capacity.
