Objective:Establish total cerebral ischemia model and Encephalo-Myo-Syna ngiosis model, explore the behavioral and histological changes to study the role of revascularization in the treatment of vascular dementiaMethod:Healthy adult Sprague-Dawley rats (n=40) were divided into blank group (n=10), treatment group (n=15) and control group (n=15). After anesthetized with 10% chloral, rats were supine fixed on the operating table, take one side and make a longitudinal incision on the anatomical location of the common carotid artery, stripping the common carotid artery, ligation of the proximal and distal ends truncation. One week later, using the same method to divide the other side of common carotid artery. The procedure of blank group is the same as operation group, except for dividing of bilateral common carotid arteries.4 weeks later, take one group of dementia SD as treatment group and perform the Encephalo-Myo-Synangiosis procedure, the other group as control group which exclude Encephalo Myo Synangiosis procedure. After anesthesia, make a U incision on temporal part, stripping temporalis, rub out skull, removal cerebral dura mater and take temporalis apply on brain mantle surface, suture skin and muscle in situ. Make stripping temporalis on the control group, but remain the skull. Evaluate the cognitive function of all groups of SD rats with Morris water maze separately before and after operation, and evaluate again when 4 weeks after EMS. Then take the SD brain pathology samples, via HE staining, observe the change of the nerve cells morphology of Hippo-campus CA3 area under an optical microscope.Result:1.Hidden Platform Test:(1)Through the comparison between dementia group and blank group, the result has significant deviation (p<0.01). (2)Through the comparison between treatment group and control group, the result has statistical significance (p<0.05). (3).Through the comparison between treatment group and blank group, the result has significant deviation (p<0.01).2. Space Exploration Test: (1)Through the comparison between dementia group and blank group, the result has significant deviation (p<0.01). (2)Through the comparison between treatment group and control group, the result has statistical significance (p<0.05). (3)Through the comparison between treatment group and blank group, the result hasn’t statistical significance (p>0.05).Conclusion:1. Establish the SD global brain ischemia model with upgrade bilateral common carotid arteries ligation is feasible.2. Apply the Encephalo-My o-Synangiosis on SD vascular dementia can obviate aggravate continue torpid ce rebral ischemia, thereby delaying the progression of dementia, to prevent further loss of cognitive function.
