Objective This study observed the effect of Chinese herbal medicine berberine on human umbilical vein endothelial cells induced by vascular endothelial growth factor in proliferation and migration,to clarify whether berberine can inhibit the proliferation and migration of HUVECs induced by VEGF, and explore the role of STAT3 in this process, so that providing the theoretical and experimental basis for the clinical application of berberine in anti-tumor angiogenesis.Methos Human umbilical vein endothelial cells line were prepared by enzyme digestion. Using Berberine,AG490(JAK signal transduction pathways specific inhibitor) to intervene the HUVECs induced by VEGF.The proliferation of the HUVECs was measured by MTT assa.The migration of the HUVECs was observed by transwell chambers.The expression levels of p-STAT3 protein were detected by Western Blot analysis.Results ① VEGF(20ng/ml) on HUVECs 24 hours, can induce HUVECs migration, proliferation, and increase p- STAT3 protein expression in parallel at the same time. ② Berberine(20μg/ml) and AG490 significantly inhibited the proliferation of HUVECs induced by VEGF, and The proliferation was inhibited by berberine in atime-dependent and dose-dependent manner( P<0.000,1). ③Berberine and AG490 significantly inhibited the migration of HUVECs induced by VEGF(P<0.01). ④Berberine significantly inhibited the p-STAT3 expression of HUVECs induced by VEGF(P<0.01).Conclusion Berberine may inhibit proliferation and migration of HUVECs induced by VEGF in vitro through the JAK2/STAT3 pathway and down regulate the expression of p-STAT3;The ingredients of Chinese medicine berberine is expected to become a good anti-tumor angiogenesis drug in clinical application In the future.
