Objective:By examining the behavior changes in Parkinson’s disease rats to acquaint the effect on the rats raised by Gui ling pa an wan;By researching the influence which raised with Gui ling pa an wan on SOD,MDA,T-AOC in Parkinson’s disease rats’ s blood serum,to discuss the possible reason why Gui ling pa an wan can treat Parkinson’s disease,provide a scientific basis for clinical medication.Methods:Fed the rats1 week,then make the rats that have no rotation behavior as experimental object,and divide them into control group and experimental group randomly.The experimental group adopts the following method to build model:(1)Anesthesia:Give the rats anesthesia by abdominal cavity injection with chloral hydrate;(2)Fix the position by stereo positioning instrument of rats:Positioning with a strict cranial flat bit;Determine the surgical site,Give rats the regular disinfection an shave the skin;Cut along the midline scalp,strip the periosteum,expose the anterior and posterior fontanel,locate the left the substantia nigra compacta,and drill through the skull to the desired location with dental drill slowly.(3)Trace injection sampler:Needle according to the determined position,and then inject 8 ug6-OHDA when arriving at the desired depth;Keep the needle retaining 30min;Pull out the needle.Cover the hole with dental adhesive,Suture the wound regularly;(4)Inject 100000u/kg penicillin for 3 days to prevent the postoperative infection.(Establish the rat model with Parkinson’s disease referencing the methods by Zhou Hou-guang and so on)Inject the sham operation rats with 0.2g/L physiological saline which contain ascorbic acid,and the rest of the conditions and process are the exactly same to the model group.Experimental group rats and sham operation group rats are fed under the same condition,Detect the behavior at the 28 th day after the surgery,using Apomorphine(APO)as derivant,and observe the behavior changes.After repeated behavior detecting,record observations for average.The successful PD rats are divided into model group,Madopar group and Gui ling pa an wan for large,medium and small dose group.Madopar group are given medicines at 26.04mg/kg/d;High-dose group are given medicines at3.75g/kg/d;Medium-group are given medicines at 1.87/kg/d;Small-dose group are given medicines at 0.93g/kg/d.All groups are fed at the same conditions.1 day 1 twice,a total of 30 days.Observe the rates behavior for many times at 3 days before building the model and the 28 th,29th,30 thdays after the mold successfully,and the 28 th,29th,30 th after fed the medicine.Record them.Kill the rats beheaded after fed the last 24 hours.Take 2ml blood,and inpour into the test.Serum the serum.Determin SOD by WST method,MDA by associates method,T-AOC by Fe3+ reduction method,all experimental steps are completed by the staff in the laboratory of Henan Traditional Chinese Medicine Hospital,and they are operated strictly following the instructions.Results:(1)30 days after the rotation,behavior of the experience group rats compared with preoperative have statistically significant(P<0.05);(2)Before the drug intervention and after the intervention 30 days,the rats’ behavior change has statistically significant(P<0.05);(3)Madopar group,Gui ling pa an wan large dose group,middle dose group and small dose group compared with model group have statistically significant at SOD vigor(P<0.05);Madopar group,Gui ling pa an wan high-dose group,middle-dose group compared with Gui ling pa an wan small-dose group have statistically significant at SOD vigor(P<0.05);(4)Madopar group,Gui ling pa an wan high-dose group compared with model group have statistically significant at MDA value(P<0.05);(5)Madopar group,Gui ling pa an wan large-dose group,middle-dose group and small-dose group compared with model group have statistically significant at T-AOC vigor;(P<0.05);Madopar group,Gui ling pa an wan large-dose group compared with Gui ling pa an wan middle-dose group and small-dose group have statistically significant at T-AOC vigor(P<0.05).Conclusion:Gui ling pa an wan can improve the rotation of the PD model rats and other abnormal behavior;Gui ling pa an wan can improve SOD activity,T-AOC activity,and reduce the MDA value in PD rats;Different doses have different effects on SOD,MDA,T-AOC;Inhibiting oxidative stress reaction may be the probablemechanisms that Gui ling pa an wan treat the Parkinson’s disease.
